Abstract :
เอนไซม์ไคโตไบเอสหรือเอ็น อะซิทิลกลูโคซามินิเดสจัดเป็นเอนไซม์ที่อยู่ในไกลโคซิลไฮโดรเสล แฟมิลี 3 หรือแฟมิลี 20 ซึ่งเอนไซม์ทั้งสองแฟมิลีมีความแตกต่างกันทั้งลำดับของกรดอะมิโนและกลไกในการเร่งปฏิกิริยา ในการศึกษาครั้งนี้ ผู้วิจัยได้ทำการแยกยีนของเอนไซม์เอ็น อะซิทิลกลูโคซามินิเดสแฟมิลี GH-20 สองตัวคือ VhNag1 และ VhNag2 จากจีโนมของแบคทีเรีย Vibrio harveyi (ชื่อเดิมคือ V. carchariae) สายพันธุ์ 650 ต่อมาได้ทำการโคลนยีนทั้งสองเข้า pQE60 exression vector ซึ่งสามารถทำการผลิตรีคอมบิแนนท์โปรตีนใน E. coli เจ้าบ้านสายพันธุ์ M15 ได้ การวิเคราะห์สายโพลีเพปไทด์ของ VhNag1 มีขนาด 88,849 Da มีค่า pI เท่ากับ 4.9 มีค่า pH optimum ในช่วงกว้างคือ 6.5-7.5 ส่วนสายโพลีเพปไทด์ของ VhNag2 polypeptide มีน้ำหนักโมเลกุลเท่ากับ 73,143 Da มีค่า pI เท่ากับ 5.4 และมีค่า pH optimum เท่ากับ 7.0 การศึกษาการย่อยสลายสับสเตรทพบว่ารีคอมบิแนนท์ VhNag1 และ VhNag2 ที่ถูกผลิตโดย E. coli สามารถย่อยไคตินสับสเตรทได้ทุกชนิด แต่ประสิทธิภาพในการย่อยสลายของเอนไซม์ VhNag2 ดีกว่าเอนไซม์ VhNag1 อย่างน้อย 10 เท่า ผลการทดสอบทางชีวเคมีและข้อมูลทางจลนพลศาสตร์พบว่าเอนไซม์ VhNag2 สามารถย่อยไคตินโอลิโกแซคคาร์ไรด์แบบเป็นลำดับและให้ผลิตผลสุดท้ายเป็น GlcNAc ซึ่งบ่งบอกว่า VhNag2 มีลักษณะเป็น exolytic enzyme นอกจากผลการเปรียบเทียบความสามารถในการย่อยสับสตเรทต่าง ๆ พบว่า VhNAg2 ย่อย pNP-GlcNAc2 ได้ดีที่สุด ส่วนเมื่อเทียบกับไคตินโอลิโกแซคคาร์ไรด์ด้วยกันพบว่าไคโตเตตระโอสเป็นสับสเตรทที่ดีที่สุดของ VhNag2 ABSTRACT Chitobiases or β-N-acetylglucosaminidases are classified into glycosyl hydrolases family 3 (GH-3) or family 20 (GH-20), both are different in sequence identity and mode of enzyme action. In this study, two genes corresponding to GH-20 GlcNAcase homologues (namely VhNag1 and VhNag2) were successfully isolated from the genome of V. harveyi (formerly classified as V. carchariae) type strain 650. Both genes were cloned into pQE50 expression vector, which could be expressed in E. coli M15 host cells. The VhNag1 polypeptide has a molecular weight of 88,849 Da, a pI of 4.9 and a broad range of pH optimum from 6.5 to 7.5, whereas the VhNag2 polypeptide has a molecular weight of 73,143 Da, a pI of 5.4 and a pH optimum of 7.0. The recombinant VhNag1 and VhNag2 expressed by E. coli were found to degrade all the tested chitin substrates with VhNag2 being at least ten fold more active than VhNag1. The results obtained from different biochemical assays and kinetic characterization consistently suggested that VhNag2 degraded chitin oligosaccharides in a sequential manner, generating GlcNAc as the final end product, which suggests that VhNag2 is an exolytic enzyme. VhNag2 acts most efficiently on pNP-GlcNAc2. For chitin oligosaccharide, chitotetraose was found to be the best substrate.
