Abstract :
บทคัดย่อ รางจืด (Thunbergi laurifolia Linn.) เป็นพืชสมุนไพรที่มีคุณสมบัติทางเภสัชวิทยาได้แก่ฤทธิ์ต้านยาฆ่าแมลงและสารเคมี ลดการติดสารเสพติด รวมทั้งแก้พิษไข้ จึงได้มีการนำไปใช้ในการแก้พิษหลายชนิดในรูปของชาชงดื่มและแคปซูลในวงการแพทย์แผนโบราณ แต่ยังไม่มีรายงานการศึกษาความเป็นพิษในรูปแบบของสารสกัดในสารละลายชนิดต่าง ๆ มาก่อนการวิจัยครั้งนี้จึงได้ศึกษาความเป็นพิษเฉียบพลันของสารสกัดรางจืดโดยการเตรียมในสารละลาย 3 ชนิดได้แก่ น้ำ เอทานอล และอะซีโตน เมื่อบริโภคขนาดสูง โดยใช้หนูขาวสายพันธุ์ Wistar ทั้งเพศผู้และเมียเป็นสัตว์ทดลองผลการทดลองพบว่าสารสกัดรางจืดในสารละลาย 3 ชนิด ขนาด 2,000 และ 15,000 มิลลิกรัมต่อน้ำหนักตัว 1 กิโลกรัม ไม่เปลี่ยนแปลงพฤติกรรมโดยทั่วไป และไม่เปลี่ยนแปลงน้ำหนักตัวของหนูขาว รวมทั้งไม่พบความผิดปกติของอวัยวะภายในทั้งหมด ในการศึกษาความเป็นพิษกึ่งระยะยาวของสารสกัดรางจืดน้ำและเอทานอล ขนาดเทียบเท่าการดื่มชาในคน (1.46 และ 1.025 มิลลิกรัมต่อกิโลกรัม ตามลำดับ) เป็นระยะเวลา 90 วัน โดยมีกลุ่มควบคุมและกลุ่มย้อนกลับ(หยุดสารสกัด และสังเกตอาการต่ออีก 14 วัน) พบว่าไม่เปลี่ยนแปลงพฤติกรรมโดยทั่วไป และน้ำหนักตัวเพิ่มขึ้นตามปกติ ส่วนน้ำหนักของอวัยวะ ปอด ตับ ม้าม อวัยวะสืบพันธุ์เพศผู้และเมีย ไม่แตกต่างทางกันทางสถิติ ยกเว้นน้ำหนักสัมพัทธ์เฉลี่ยของหัวใจ และไต มีแนวโน้มมีขนาดเล็กลง เมื่อเปรียบเทียบกับกลุ่มควบคุม (p<0.05) ส่วนค่าทางเคมีคลินิค (RBC, WBC, Hemoglobin, hematocrit, PMN, PCV, MCV, MCH, MCHC, RDW,MPV) ค่าเลือดทางโลหิตวิทยา (BUN, creatinine, albumin) และเอนไซม์ในตับ (AST, ALT, ALP) พบว่าค่าทางเคมีคลินิคและค่าเอนไซม์ในตับไม่มีความแตกต่างกันทางสถิติ ยกเว้นกลุ่มเพศผู้ที่ได้รับสารสกัดรางจืดเอทานอล พบว่า มีค่าเฉลี่ยระดับ ALT สูงขึ้น เท่ากับ 41.53±24.65 U/L จากกลุ่มควบคุมเท่ากับ 32.40±6.65 U/L อย่างมีนัยสำคัญทางสถิติ (p<0.05) และกลุ่มศึกษาผลย้อนกลับที่ได้รับสารสกัดรางจืดเอทานอลในหนูเพศผู้ พบว่า มีค่าเฉลี่ยของระดับ AST มีค่าลดลง เท่ากับ 108.20± 15.80 U/L จากกลุ่มควบคุมเท่ากับ 156.00±64.85 U/L อย่างมีนัยสำคัญทางสถิติ (p<0.05) และค่า creatinine กลุ่มย้อนกลับที่ได้รับสารสกัดรางจืดน้ำในเพศเมียมีค่าเพิ่มขึ้นจากกลุ่มควบคุม 0.89±0.14 อย่างมีนัยสำคัญยิ่งทางสถิติ ส่วนการทดสอบความเป็นพิษต่อเซลล์ โดยวัดระดับของ malondiadehyde (MDA) พบว่าในเพศผู้กลุ่มทดสอบที่ได้รับน้ำมีค่าลดลงมากกว่ากลุ่มควบคุมอย่างมีนัยสำคัญยิ่งทางสถิติ (p<0.05) คำสำคัญ: สารสกัดรางจืด ความเป็นพิษกึ่งระยะยาว หนูขาว Abstract Rang Chuet (Thunbergia laurifolia Linn.) is Thai medicinal plant with pharmacological properties, such as antidote for many toxic substances in pesticide and others, drug addicted reduction, and decrease body temperature. This study investigated sub-chronic toxicity of Rang Chuet extract for safety concern in human use.In the determine acute toxicity studies of Rang Chuet in rats at 2,000 and 15,000 mg per kg body weight of three Rang Chuet extracts by using different solvents including water, ethanol, and acetone extraction. An acute toxicity study was performed by using both sexes of Wistar rats. The results revealed that all three types of Rang Chuet leave extract at a single dose of 2,000 mg per kg bodyweight did not alter the general behavior and the feature of the visceral organs of rats. In addition, the extracts with oral dose of 15,000 mg per kg bodyweight did not produce mortal or change in gross morphology of the internal organs of the rats, as well as no histological changes of visceral organs in every group were found.In sub-chronic toxicity study was performed using Wistar rats of both sexes (5 of each sex in each group) were gavarged with suspension of ethanol and water Rang Chuet extracts at dose 1.025 and 1.46 mg/body weight/day, respectively (quality of consumer drink for three times per day ) for 90 days. In the satellite group were given the suspension at the same dose for 90 days and observed thereafter 14 days in order to study the reversibility of adverse effects.The result of study showed treated ethanol and water Rang chuet extracts were no significant effect on average body weight, relative organ weight(heart, lungs, liver, kidneys, adrenal gland, spleen, testis or ovary), histopathology of organ, clinical biochemistry(blood urea nitrogen, albumin, creatinine), hematological parameters(RBC, WBC, hemaglobin, hematocrit, PMN, PCV, MCV, MCH, MCHC, RDW, MPV) and liver enzymes (alkaline phosphate, alanine aminotransferase, aspartate aminotransferase) except, increase ALT in male rat treated ethanol Rang chuet extracts group (p<0.05) and decrease AST in male treated ethanol Rang chuet extracts satellite group and increase Creatinine in female treated water Rang chuet extracts satellite group. In addition, malondialdehyde assay, a by product of lipid peroxidation, when compared with control group showed decrease in male rat treated water Rang chuet extract group (p<0.01).
