Abstract :
การศึกษาความเป็นไปได้ของการเก็บรักษาน้ำเชื้อปลากระโห้โดยวิธีการแช่แข็ง แบ่งการศึกษาออกเป็น 3 การทดลองดังนี้ 1) ผลของสาร extender ชนิดและระดับความเข้มข้นของสาร cryoprotectant ที่เหมาะสมสำหรับการเก็บรักษาน้ำเชื้อปลากระโห้แบบแช่แข็ง 2) ผลของอัตราการลดอุณหภูมิ ที่เหมาะสมสำหรับการเก็บรักษาน้ำเชื้อปลากระโห้แบบแช่แข็ง และ 3) ผลของขนาดและภาชนะบรรจุ ที่เหมาะสมสำหรับการเก็บรักษาน้ำเชื้อปลากระโห้แบบแช่แข็ง สำหรับการทดลองที่ 1 มีการใช้สาร extenders 2 ชนิด (Hanks balanced salt solution-HBSS, และ modified cortland solution-MC) ร่วมกับสาร cryoprotectant 3 ชนิด (dimethyl sulfoxide-DMSO, dimethyl acetamide-DMA และ propylene glycol- PG) ที่ระดับความเข้มข้น 5, 7.5 และ 10% สำหรับ DMSO และ DMA และที่ระดับความเข้มข้น 10, 15 และ 20% สำหรับ PG เก็บน้ำเชื้อเป็นเวลา 2 วันจากนั้นนำน้ำเชื้อแช่แข็งไปประเมินคุณภาพจากอัตราการเคลื่อนที่ อัตราการมีชีวิต และอัตราการปฏิสนธิ พบว่าเมื่อใช้ 10% DMSO เป็นสาร cryoprotectant ร่วมกับ MC ให้ผลอัตราการเคลื่อนที่ และอัตราการปฏิสนธิสูงกว่าทรีตเมนต์อื่นๆ (P<0.05) อีกทั้งยังให้อัตราการเคลื่อนที่ไม่แตกต่างกับการใช้น้ำเชื้อสด สำหรับการใช้ DMA เป็นสาร cryoprotectant พบว่าเมื่อใช้ 10% DMA ร่วมกับ HBSS ให้ผลอัตราการปฏิสนธิสูงกว่าทรีตเมนต์อื่น ๆ อย่างไรก็ตามการใช้ DMSO เป็นสาร cryoprotectant ให้อัตราการปฏิสนธิสูงกว่าเมื่อเปรียบเทียบกับชุดการทดลองที่มีการใช้ DMA เป็นสาร cryoprotectant สำหรับการใช้ PG ให้ผลอัตราการปฏิสนธิต่ำในทุกทรีตเมนต์ การทดลองที่ 2 ศึกษาผลของอัตราการลดอุณหภูมิ ที่เหมาะสมสำหรับการเก็บรักษาน้ำเชื้อปลากระโห้แบบแช่แข็ง โดยเลือกสาร extender และ cryoprotectant ที่ให้อัตราการปฏิสนธิดีที่สุดจากการทดลองที่ 1 คือ 10% DMSO ร่วมกับ MC และ 10% DMA ร่วมกับ HBSS มาทำการศึกษาอัตราการลดอุณหภูมิ โดยใช้อัตราการลดอุณหภูมิแบบ One-step และ Two-steps ร่วมกับอุณหภูมิสุดท้าย (Endpoint) 3 ระดับ (-40, -60 และ -80 ๐C) พบว่าการใช้ 10% DMSO ร่วมกับ MC ให้อัตราการปฏิสนธิสูงกว่าการใช้ 10% DMA ร่วมกับ HBSS (P<0.05) และในการทดลองที่ 3 ศึกษาผลของขนาดและภาชนะบรรจุที่เหมาะสมสำหรับการเก็บรักษาน้ำเชื้อปลากระโห้โดยวิธีแช่แข็งโดยการนำทรีตเมนต์ที่ให้อัตราการปฏิสนธิดีที่สุดจากการทดลองที่ 2 คือ 10% DMSO ร่วมกับ MC และใช้อัตราการลดอุณหภูมิแบบ One-step ณ จุด end point -40 ๐C มาทำการศึกษาขนาดของภาชนะบรรจุ 5 ขนาด คือ (0.25, 0.5, 1, 2 และ 5 มิลลิลิตร) พบว่าการใช้ภาชนะบรรจุแบบ Straw ขนาด 0.25, 0.5 และ 1 ml และการใช้ cryovial ขนาด 2 และ 5 ml ไม่มีผลต่ออัตราการเคลื่อนที่ อัตราการมีชีวิต และอัตราการปฏิสนธิ (P>0.05) ABSTRACT This present study examined the feasibility of cryopreservation of Giant barb, Catlocarpio siamensis sperm. Three major experiments were carried out: 1) the effects of extenders and cryoprotectants 2) the effects of freezing rates and 3) the effect of sizes and types of containers on the cryopreservation of C. microlepis sperm were investigated. In the first experiment, effect of two extenders (Hanks balanced salt solution-HBSS and modified cortland solution-MC) with three cryoprotectants (dimethyl sulfoxide-DMSO, dimethyl acetamide-DMA and propylene glycol- PG) at concentration of 5, 7.5 และ 10% for DMSO and DMA, and 10, 15, 20% for PG on the cryopreservation of C. microlepis sperm were investigated. Sperm were stored for 48 h in a liquid nitrogen container (-196 °C). They were then airthawed at room temperature (25 °C), and motility, viability and fertilization rates were assessed. The higher motility rates and fertilization rates (P<0.05) were achieved with the combination of 10% DMSO+MC. In addition, motility percentage was not difference with the control (fresh sperm). The highest fertilization rate of 12.88±1.10% (41% of fresh sperm) was resulting from10% DMA+HBSS. Among cryoprotectants used, DMSO as cryoprotectant yielded a higher fertilization rates compared to DMA or PG. The highest fertilization rate from each combination of extender and cryoprotectant from the first study (10% DMSO+MC and 10% DMA+HBSS) were used to investigate the effect of freezing rates (one-step and two-steps) with end point at -40, -60 and -80°C on the cryopreservation of C. microlepis sperm (experiment 2). Among treatments used the combination of 10% DMSO+MC yielded a higher motility, viability and fertilization rates than 10% DMA+HBSS. In the third experiment, the effect of sizes and types of the containers by using 10% DMSO+MC with one-step freezing rate at end point -40°C were determined. The results showed that sizes and types of the containers used did not affect fertilization, motility and viability percentages (P>0.05).
