บทคัดย่อภาษาไทย Von Willebrand factor (VWF) เป็น glycoprotein ขนาดใหญ่ที่อยู่ในพลาสมามีบทบาทสำคัญในกระบวนการทำให้เลือดหยุดและการเกิดลิ่มเลือดอุดตัน โดยไปส่งเสริมให้เกล็ดเลือดเกาะกับชั้นของ endothelium ในบริเวณหลอดเลือดที่ได้รับความเสียหาย VWF แต่ละโมโนเมอร์ประกอบด้วย domain 4 แบบ (A ถึง D) จำนวน 13 domain มาเรียงต่อกัน A1 domain ของ VWF เป็นโครงสร้างที่มีบทบาทสำคัญในการเริ่มต้นปฏิกิริยาการเกาะตัวกันของเกล็ดเลือด ด้วยเหตุนี้ความสัมพันธ์ระหว่างโครงสร้างและหน้าที่ของ A1 domain จึงเป็นการศึกษาที่ได้รับความสนใจเป็นอย่างมากทั้งในแง่ของความเข้าใจหน้าที่ของ domain นี้ใน in vivo และในแง่ของการพัฒนายาชนิดใหม่ ๆ ที่มีฤทธิ์ต้านการเกิดลิ่มเลือดอุดตัน การผลิต recombinant A1 domain ขึ้นจึงมีคุณค่าอย่างมากในการเป็นเครื่องมือที่ใช้ศึกษาโครงสร้างและหน้าที่ของโปรตีนชนิดนี้ ในการศึกษาวิจัยนี้ ผู้วิจัยได้ทำการ clone ยีนของ VWF A1 domain จาก genomic DNA ของคนแทนการ clone ยีนจาก mRNA ซึ่งวิธีที่ค้นพบนี้มีข้อได้เปรียบ คือ สามารถนำไปใช้ศึกษาโครงสร้างและหน้าที่ของ A1 domain ในผู้ที่มี polymorphism ของ VWF gene ต่างกันได้ ในเบื้องต้นผู้วิจัยได้พยายามผลิต recombinant A1 domain จากเซลล์ของสัตว์เลี้ยงลูกด้วยนมเพื่อโปรตีนที่ผลิตขึ้นจะมีส่วนที่เป็น glycosylation เช่นเดียวกับโปรตีนในธรรมชาติของคน แต่ทว่าโปรตีนที่ผลิตได้จากการใช้เซลล์ COS-7 มีปัญหาด้านการละลายทำนองเดียวกับโปรตีนส่วนใหญ่ที่เคยมีรายงานมาก่อนหน้านี้แล้ว อย่างไรก็ตามผู้วิจัยประสบความสำเร็จในการสร้าง recombinant A1 domain ที่สามารถละลายน้ำได้จากแบคทีเรีย โดยการออกแบบให้มีการติด hexahistidine tag ทางด้าน C-terminal ของสายโปรตีนซึ่งเป็นอีกด้านหนึ่งของ A1 domain ซึ่งเกิดจากการสร้างพันธะ disulfide ระหว่าง Cys1272 Cys1458 วิธีนี้มีความแตกต่างจากการศึกษาทุกการศึกษาที่เคยมีรายงานไปแล้วคือ สามารถสกัดแยก recombinant A1 domain โดยโปรตีนที่สร้างขึ้นยังอยู่ในสภาพเดิมตามธรรมชาติ ไม่มีการทำลายพันธะ non-covalent หรือพันธะ disulfide แต่อย่างใด จากการศึกษาด้านโครงสร้างโดยการใช้ recombinant A1 ที่สร้างขึ้นนี้ ผลการทดลองแสดงให้เห็นว่าโครงสร้างของ A1 domain เกิดการเปลี่ยนแปลงหลังจากบ่มด้วย ristocetin โดยการใช้ SDS-PAGE และ HPLC-MS ในการวิเคราะห์ ข้อมูลที่ได้จากการศึกษานี้เป็นหลักฐานสำคัญเพิ่มเติมที่แสดงให้เห็นว่า ristocetin ชักนำให้เกิดการเปลี่ยนแปลงโครงสร้างของ VWF A1 domain ระหว่างที่เกิดการเกาะตัวกันของเกล็ดเลือด คำสำคัญ: วอนวิลลิแบรนด์แฟคเตอร์, เอวันโดเมน, รีคอมบิแนนท์, ริสโทซีติน, โครงสร้าง Abstract Von Willebrand factor (VWF) is a multimeric, plasma glycoprotein that plays an essential role in hemostasis and thrombosis. It promotes platelet adhesion to damaged vascular endothelium. The VWF monomer includes 13 domains, which are multiples of four domain types (A to D). The A1 domain in VWF domain contains multiple binding sites with critical roles in the initiation of platelet aggregation. The relationship of structure to function in the VWF A1 domain is a topic of intense interest, both from the perspective of understanding how the function of this domain is regulated in vivo and from the standpoint of developing novel antithrombotic agents. An isolated A1 domain expressed in recombinant form can serve as an invaluable tool for studying its structural and functional attributes. In this study, the investigator has cloned A1 domain of human VWF from genomic DNA, rather than from mRNA. This approach has some advantages, which can facilitate subsequent studies of the structural and functional consequences of specific polymorphisms or mutations in the VWF gene. First, a recombinant A1 domain was generated from the mammalian cells so that the protein produced would have glycosylation parts as native protein from human. The protein expressed in COS-7 cells was encountered problems with solubility similar to those from many previous reports. However, a soluble form of the recombinant A1 domain of VWF was successfully generated from bacteria. This was accomplished through attachment of a C-terminal hexahistidine tag and judicious choice of the extent of flanking regions on either side of the critical Cys1272 Cys1458 disulfide bond. This strategy permits extraction and purification of the recombinant A1 domain protein entirely under native conditions, thereby preserving its non-covalent and disulfide bonds. In the conformation study, the results showed that structure of the recombinant A1 domain was changed after incubation of ristocetin determined by SDS-PAGE and HPLC-MS. This data provides additional important evidence that conformational changes of the A1 domain occur during the induction of platelet aggregation by ristocetin.
