Project Project Title : การแยกยีนไคติเนสจากเชื้อแบคทีเรียในทะเล Vibrio alginolyticus สายพันธุ์ 283 (Isolation of a Chitinase from a Marine Bacterium, Vibrio alginolyticus 283)     downloaded 69 times Researcher Name : วิภา สุจินต์ (Wipa Suginta) Abstract : บทคัดย่อ ในงานวิจัยชิ้นนี้ผู้วิจัยได้ทำการสกัดยีนไคติเนสจากจีโนมของเชื้อแบคทีเรียในทะเล Vibrio alginolyticus สายพันธุ์ 283 โดยทำการเตรียมชิ้นของดีเอ็นเอด้วยการตัดด้วยเอนไซม์ตัดจำเพาะ Sau3IA ขนาด 3-8 kb และตรวจหายีนไคติเนสโดยวิธีทางอิมมูโนวิทยาโดยใช้ anti-V. carchariae chitinase polyclonal antibodies เป็นตัวตรวจจับ การวิเคราะห์การแสดงออกของยีนไคติเนส ด้วย SDS-PAGE และ immunoblotting ของชิ้นดีเอ็นเอที่นำเข้าสู่พลาสมิด pBluescript II KS(-) และแบคทีเรีย DH5 พบว่าโคลน GC1–GC6 ที่ให้ผลบวกกับแอนติบอดีมีชิ้นส่วนของดีเอ็นเอที่มียีนไคติเนสเป็นองค์ประกอบ การทำแผนที่ดีเอ็นเอด้วยการตัดด้วยเอนไซม์ตัดจำเพาะหลาย ๆ ชนิด พบว่าชิ้นดีเอ็นเอของโคลน GC6 มีขนาดประมาณ 10 กิโลเบส และพบว่าการตัดชิ้นดีเอ็นเอดังกล่าวด้วยเอนไซม์ KpnI ทำให้ได้ชิ้นดีเอ็นเอขนาดเล็กลงเหลือ 7 กิโลเบส การวิเคราะห์โปรตีนด้วย SDS- PAGE immunoblotting และการหาแอคติวิตี้โดยใช้ไกลคอลไคติน (glycol-chitin) เป็นสับสเตรทแสดงให้เห็นว่ายีนไคติเนสที่สกัดได้สามารถสร้างโปรตีนขนาด 63 กิโลดัลตัน การวิเคราะห์หาลำดับนิวคลิโอไทด์บางส่วนพบว่า ลำดับนิวคลิโอไทด์ชิ้นดีเอ็นเอของโคลน GC6 ที่ย่อยด้วย KpnI มีความเหมือนกับลำดับนิวคลิโอไทด์ของยีนไคติเนสเอ ของเชื้อแบคทีเรีย V. carchariae คือ 98.3% Abstract In this research we describe isolation of a chitinase gene from genomic DNA of a marine bacterium, Vibrio alginolyticus strain 283. The 3-8 kb DNA sizes were prepared by digesting with Sau3AI and a chitinase gene was detected immunologically using anti-V. carchariae chitinase polyclonal antibodies as a probe. SDS –PAGE and immunoblotting of all the generated DNA fragments, which were cloned into the pBluescript II KS (-) plasmid and transformed into DH5 host cells, suggested that clones GC1-GC6 carried a DNA insert containing a chitinase gene. Analysis of DNA mapping with different restriction enzymes suggested that clone GC6 carried a DNA insert of approx. 10 kb. This insert was reduced to 7 kb when digested with KpnI. As revealed by SDS-PAGE, immunoblotting, and chitinase assay on native PAGE using glycol-chitin as substrate , the KpnI-digested DNA could express a 63-kDa protein, which highly corresponded to chitinase A isolated from V. carchariae. Partial nucleotide sequencing showed that nucleotide sequence of the isolated gene was 98.3% identical to that of V. carchariae chitinase A.
