Abstract :
บทคัดย่อ จากการคัดเลือกแบคทีเรียกลุ่มที่คาดว่าอาศัยในเนื้อเยื่อของต้นข้าว ที่มีความสามารถในการตรึงไนโตรเจน โดยใช้อาหารเลี้ยงเชื้อ Rennie medium (RMR) ที่ไม่ใส่แหล่งอาหารไนโตรเจน จำนวนทั้งสิ้น 256 ไอโซเลท จากต้นข้าวที่ทำการปลูกในจังหวัดพระนครศรีอยุธยา นครสวรรค์ เชียงใหม่ และจังหวัดนครราชสีมา พบว่าจำนวนประชากรของแบคทีเรียที่พบส่วนใหญ่ได้แก่ ที่ใบและราก ซึ่งมีจำนวนประชากรอยู่ในช่วง 106 เซลล์/น้ำหนักสด (กรัม) ส่วนที่ลำต้นมีเพียง 104-105 เซลล์/ น้ำหนักสด (กรัม) ประชากรส่วนใหญ่เป็นแบคทีเรียแกรมบวกและมีรูปร่างแบบแท่ง การวิเคราะห์ทางชีวเคมีแสดงให้เห็นว่าแบคทีเรียส่วนใหญ่ไม่มีความสามารถในการใช้มาเลทเป็นแหล่งอาหารคาร์บอน แต่สามารถใช้แมนนิทอล, กลูโคส และอะราบิโนส เป็นแหล่งอาหารคาร์บอนได้ นอกจากนี้ยังพบว่าสามารถผลิตเอ็นไซม์เซลลูเลสและเพคติเนสด้วย การศึกษาครั้งนี้ได้เลือก 5 สายพันธุ์ ซึ่งมีความสามารถในการตรึงไนโตรเจนสูงมาทำการศึกษาต่อ โดยกลุ่มที่มีความสามารถในการตรึงไนโตรเจนอยู่ในช่วง 66-753 นาโนโมลเอซิลิน ต่อจำนวนประชากรที่มีชีวิต 106 เซลล์ พบว่ามี 3 สายพันธุ์จากที่คัดเลือก 4 สายพันธุ์ ที่สามารถผลิต Indole Acetic Acid (IAA) จากนั้นทำการหาลำดับเบสของยีน 16S rRNA บางส่วนของสายพันธุ์ดังกล่าว พบว่ามีความใกล้เคียงกับแบคทีเรีย Bacillus subtilis (Acc. No. AF391127), Azospirillum sp. (Acc. No. AB049109), low G + C gram-positive (Acc. No. AB002342) และ B. licheniformis (Acc. No. NC000964.1) นอกจากนี้ยังยืนยันผลการแสดงออกของสายพันธุ์ดังกล่าวในเนื้อเยื่อข้าวโดยใช้ Green Fluoresent Protein (GFP) และ gus เป็น reporter gene ด้วยเทคนิค electroporation แล้วนำไป inoculate ร่วมกับการปลูกข้าวในสภาวะปลอดเชื้อ พบว่าแบคทีเรียที่มีความใกล้ชิดกับ B. licheniformis สามารถเข้าอาศัยในเนื้อเยื่อของต้นข้าวได้จริง นอกจากนี้ ทำการทดสอบยืนยันโดยสำรวจด้วยกล้องจุลทรรศน์อิเลคตรอนแบบส่องกราด พบว่าผลเป็นไปในทำนองเดียวกัน และยังพบพัฒนาการทางสัณฐานวิทยาต่าง ๆ กันไปตามช่วงอายุของต้นข้าวด้วย Abstract Isolation of putative N2-fixing endophytic bacteria from rice was conducted on basis of culturing on Rennie-N-free medium (RMR). Total 256 bacterial isolates were obtained from rice specimens collected from Ayudhaya, Nakhon-Sawan, Chieng Mai and Nakhon Ratchasima. The main number population of bacteria was established in root and leaf approximately 106 cells/g fresh weight. Whilst, few population number in the range between 104-105 cells/g fresh weight was found in the stem. Most of bacterial isolates were gram-positive and rod-shaped. The carbon source utilization indicated that most of the isolates cpuld not to use malate as sole carbon source. However, other sources of carbon as manitol, glucose and arabinose could be utilized. In addition, cellulase and pectinase enzymes production was also be detected. The top five bacterial isolate have high efficiency in N2-fixation were selected. The range of N2-fixing efficiency was in the range 66-753 nMC2H4/106 cells. Three of four isolates showed IAA production capability. From partical 16S rRNA gene analysis, they have similarity with Bacillus subtilis (Acc. No. AF391127), Azospirillum sp. (Acc. No. AB04910), Low G+C gram-positive bacteria (Acc. No. AB002342) and B. licheniformis (Acc. No. NC 000964.1). In addition, to confirm establishment of bacterial isolates in rice tissue, electroporation of green fluorescent protein (GFP) and gus gene was carried out prior to inoculate to rice. Only bacterial isolate which has similarity with B. licheniformis showed endophytic property. Cell differentiation of this strain inside the rice tissue was also elucidated by scaning electron microscope. Its different morphological changes were observed along with the rice age.
