บทคัดย่อ การทดลองนี้ได้ทำการโคลนนิ่งโดยใช้เซลล์ไฟโบรบลาสจากผิวหนังหน้าท้องแมวดาวเพศเมียเป็นเซลล์ต้นแบบฉีดเข้าไปในไข่แมวบ้านที่ดูดสารพันธุกรรมออกแล้ว โดยทำการทดลองเปรียบเทียบกับการโคลนนิ่งตัวอ่อนแมวบ้านโดยใช้เซลล์ไฟโบรบลาสจากใบหูเป็นเซลล์ต้นแบบและตัวอ่อน Parthenogenetic ในขบวนการโคลนนิ่งพบว่าอัตราการเชื่อมติดของเซลล์ผิวหนังหน้าท้องแมวดาวสูงกว่าเซลล์แมวบ้าน (84.5% และ63.3%) หลังจากนั้นแบ่งตัวอ่อนออกเป็นสองกลุ่ม กลุ่มแรกกระตุ้นด้วย 7% เอทานอล นาน 5 นาทีและกระตุ้นต่อด้วย CHX-CD นาน 5 ชั่วโมง ส่วนกลุ่มที่สองกระตุ้นด้วย CHX-CD นาน 5 ชั่วโมง จากนั้นเลี้ยงตัวอ่อนทั้งหมดในหลอดแก้วเพื่อตรวจสอบการแบ่งตัวและการเจริญสู่ระยะบลาสโตซีส พบว่า ในกลุ่มแรก ตัวอ่อนแมวดาวมีอัตราการแบ่งตัวต่ำกว่าตัวอ่อนแมวบ้านและตัวอ่อน Parthenogenetic ในขณะที่ตัวอ่อนแมวบ้านและตัวอ่อน Parthenogenetic มีอัตราการแบ่งตัวใกล้เคียงกัน แต่อัตราการเจริญสู่ระยะมอรูลาและ บลาสโตซีส ของตัวอ่อน Parthenogenetic สูงกว่าตัวอ่อนแมวดาวแต่ใกล้เคียงกับตัวอ่อนแมวบ้านโคลนนิ่ง (59.7%, 12.0%, 35.5%, 9.2% และ 40.0%, 11.2% ตามลำดับ) ในกลุ่มที่สอง พบว่าอัตราการแบ่งตัวของตัวอ่อนทั้งสามชนิดใกล้เคียงกัน แต่การเจริญสู่ระยะมอรูลาและบลาสโตซีสของตัวอ่อน Parthenogenetic มีสูงกว่าตัวอ่อนแมวดาวและแมวบ้านโคลนนิ่ง (43.3%, 16.4%; 37.5%, 7.9% และ 29.3%, 5.2% ตามลำดับ) จากการทดลองสรุปได้ว่าตัวอ่อนแมวดาวโคลนนิ่งข้ามสปีชีย์โดยใช้ไข่แมวบ้านเป็นไซโตพลาสซึมผู้รับสามารถเจริญจนถึงระยะบลาสโตซีสได้และวิธีการกระตุ้นด้วย 7% เอทานอล นาน 5 นาทีและกระตุ้นต่อด้วย CHX-CD นาน 5 ชั่วโมง เป็นวิธีที่ทำให้ได้ตัวอ่อนระยะบลาสโตซีสมากกว่ากลุ่มที่กระตุ้นด้วย CHX-CD นาน 5 ชั่วโมง Abstract In this study, we performed cloning of leopard cat using fibroblast cells from abdominal skin compared with ear fibroblast of domestic cat and parthenogenetic activation. From the experiments, we found that the fusion rate between leopard cat fibroblast and enucleated domestic cat oocytes was higher than domestic cat cells (84.5% and 63.3% respectively). Both groups of the reconstructed embryos were separated into 2 groups. The first group was activated by 7% ethanol for 5 minutes then followed by CHX-CD for 5 h. The second group was directly cultured in CHX-CD for 5 h. (without ethanol). Then, all of the activated embryos were cultured in vitro to evaluate the cleavage rates and embryo developmental rates. After fist group activation, we found that the cleavage of leopard cat embryos were lower than domestic cat embryos and parthenogenetic embryos as well. While the developmental rates of domestic cat embryos and parthenogenetic embryos were similar. However, the development and blastocyst stage of parthenogenetic embryos were higher than domestic cat embryos but similar to leopard cat embryos (59.7%, 12.0%, 35.5%, 9.2% and 40.0%, 11.2%, respectively). For the second group, the cleavage rates of leopard cat, domestic cat and parthenogenetic embryos were similar. However, the development and blastocyst stage of parthenogenetic embryos were higher than leopard cat and domestic cat embryos (43.3%, 16.4%; 37.5%, 7.9% and 29.3%, 5.2%, respectively). The results from this experiment can be concluded that leopard cat could be cloned by using domestic cat oocytes as recipient cytoplast and the suitable activation procedures for cloned leopard cat embryos is 7% ethanol for 5 min then followed by CHX-CD for 5 h.
