บทคัดย่อ จากการศึกษาผลของสาร Cryoprotectants และอัตราการลดอุณหภูมิ (freezing rates) ต่อการเก็บรักษาน้ำเชื้อปลาสวายโดยวิธีการแช่แข็งโดยเปรียบเทียบการใช้สาร cryoprotectant เพียงชนิดเดียว (dimethyl sulfoxide-DMSO, dimethyl acetamide-DMA และ methanol-MeOH หรือใช้ combination cryoprotectants โดยมี 0.9% NaCl เป็นสาร extenders และอัตราการลดอุณหภูมิที่ 10 °C min-1 จากนั้นเลือกทรีตเมนต์ที่ให้อัตราการปฏิสนธิไม่แตกต่างกับกลุ่มควบคุมในปลาสวายมาประยุกต์ใช้ในการเก็บรักษาน้ำเชื้อปลาเทโพ และศึกษาอัตราการลดอุณหภูมิที่เหมาะสมสำหรับปลาทั้ง 2 ชนิดดังกล่าว โดยใช้ French straw ขนาด 250 µl เป็นตัวเก็บรักษาน้ำเชื้อ และมี Freezer control (CL 3300) และ Cryogenesis Version 4 เป็นตัวควบคุมการลดอุณหภูมิระหว่างขบวนการแช่แข็ง นำน้ำเชื้อปลาเก็บรักษาในไนโตรเจนเหลว (-196 °C) เป็นเวลา 1 สับดาห์ จากนั้นนำน้ำเชื้อปลาแช่แข็งมาละลายที่อุณหภูมิห้อง เพื่อศึกษาประสิทธิภาพของสาร cryoprotectants และอัตราการลดอุณหภูมิที่เหมาะสมสำหรับการเก็บรักษาน้ำเชื้อปลาแบบแช่แข็ง ผลการศึกษาพบว่า combination cryoprotectants ระหว่าง10%DMSO+20%DMA, 10%DMSO+10%DMA และ20%DMA+5%MeOH ให้เปอร์เซ็นต์การปฏิสนธิในปลาสวายไม่แตกต่างกับการใช้น้ำเชื้อสด (p>0.05) และเมื่อนำมาประยุกต์ใช้ในการเก็บรักษาน้ำเชื้อปลาเทโพ พบว่า Combination cryoprotectants ในแต่ละทรีตเมนต์ ดังกล่าวนั้นให้เปอร์เซ็นต์การปฏิสนธิไม่แตกต่างกัน แต่ต่ำและแตกต่างจากน้ำเชื้อสด (P<0.05) และพบว่าการลดอุณหภูมิที่ 5 และ 10 °C min-1 มีผลให้อัตราการปฏิสนธิไม่แตกต่างกับกลุ่มควบคุม (น้ำเชื้อสด; p>0.05) สำหรับปลาสวาย ส่วนในปลาเทโพนั้นพบว่าการลดอุณหภูมิที่ 5 ºC min-1 โดยมี 10% DMSO+ 10% DMA เป็นสาร Cryoprotectant ให้อัตราการปฏิสนธิไม่แตกต่างกับกลุ่มควบคุมและสูงกว่าและแตกต่างอย่างมีนัยสำคัญทางสถิติจากการลดอุณหภูมิที่ระดับอื่นๆ (10, 20 และ 40 °C min-1, P<0.05) จากงานวิจัยในครั้งนี้สรุปได้ว่าการใช้ combination cryoprotectants ร่วมกันมากกว่าหนึ่งชนิด ในขบวนการเก็บรักษาน้ำเชื้อปลาสวายแบบแช่แข็งนั้นสามารถเพิ่มอัตราการปฏิสนธิให้สูงกว่าการใช้ Cryoprotectant เพียงชนิดเดียว นอกจากนี้พบว่าเทคนิคและวิธีการ การเก็บรักษาน้ำเชื้อปลาสวาย สามารถไปประยุกต์ใช้กับการเก็บรักษาน้ำเชื้อปลาเทโพ ซึ่งเป็นปลาในกลุ่มเดียวกันได้ และพบว่าเมื่อเพิ่มอัตราการลดอุณหภูมิเป็น 40 °C min-1 นั้นมีผลทำให้อัตราการปฏิสนธิในปลาทั้งสองชนิดมีแนวโน้มลดลง Abstract This present study examined the feasibility of cryopreservation of striped catfish, Pangasinodon hypophthalmus sperm. Two studies were carried out: (1) the effects of cryoprotectants and combination cryoprotectants on cryopreservation of P. hypophthalmus sperm and (2) the excellent treatments combination cryoprotectants from the study one, which showed high fertilization rates and did not difference from the control on the cryopreservation of P. hypophthalmus were applied to cryopreserve P. larnaudii sperm and investigate the effect of freezing rates on cryopreservation of both species. A controlled freezer (CL 3300) and Cryogenesis, version 4 was used to regulate the rate of freezing. Sperm were frozen using French straw (250 L) and stored for one week in a liquid nitrogen container. They were then air thawed at room temperature, and the effect of cryoprotectants and freezing rates on cryopreservation of P. hypophthalmus and P. larnaudii sperm were assessed. The fertilization rates of striped catfish sperm achieved with these three combination cryoprotectants (10% DMSO + 10% DMA, 10% DMSO + 20% DMA or 20%DMA + 5%MeOH) were not significantly different from the control (fresh sperm), (p>0.05). These treatments were applied to cryopreserve P. larnaudii and found that fertilization rates among three treatments were no significant difference p>0.05, but the fertilization rate was lower than the control. Fertilization rates of P. hypophthalmus, resulting from 5 and 10 °C min-1 were not significantly different from the control (fresh sperm), p>0.05. Similar result to the P. larnaudii, which showed that at 5 °Cmin-1 of freezing rate with the combination cryoprotectants of 10% DMSO + 10% DMA resulted in higher fertilization rate compared to the other freezing rates 10, 20 and 40 °C min-1 (p<0.05). This result was not significantly different from the control (p>0.05). The results from this study indicated that combination cryoprotectants used can be improved fertilization rates of Pangasiids fish. In addition, increasing freezing rate up to 40 °C min-1 resulted in decrease of fertilizing ability of both species.
