Project Project Title : การใช้เอนไซม์จากปลาและกล้าเชื้อแบคทีเรียในการหมักน้ำปลา(The Use of endogenous proteinases and bacterial starter cultures for fish sauce fermentation)     downloaded 43 times Researcher Name : จิรวัฒน์ ยงสวัสดิกุล (Jirawat Yongsawatdigul) Abstract : บทคัดย่อ วัตถุประสงค์โดยรวมของงานวิจัยนี้คือพัฒนากระบวนการผลิตที่สามารถลดระยะเวลาการหมักน้ำปลาโดยใช้เทคโนโลยีกล้าเชื้อร่วมกับเอนไซม์ทางการค้าและเอนไซม์ในตัวปลา กิจกรรมที่เหมาะสมต่อการย่อยสลายตัวเองของปลากะตัก (Stolephorus spp.) เมื่อพิจารณาจากระดับการย่อยสลายโปรตีนและกิจกรรมโปรติเนสในปลา คือที่อุณหภูมิ 50 องศาเซลเซียส เกลือโซเดียมคลอไรด์ 10% เป็นเวลา 4 ชั่วโมง โปรติเนสที่คล้ายทริปซิน (Trypsin-like proteinases) เป็นโปรติเนสกลุ่มหลักที่มีบทบาทสำคัญต่อการย่อยสลายตัวเองของปลากะตัก แบคทีเรียกรดแล็กติก 2 ไอโซเลท คือ MS33 และ MCD10-5-10 ที่คัดแยกได้จากตัวอย่างน้ำปลาหมักเดือนที่ 1 และ 5 มีรูปร่างเซลล์กลม เรียงตัวเป็นคู่ (Pairs) และแบบสี่เซลล์ (Tetrads) สามารถเจริญได้ที่ความเข้มข้นของเกลือโซเดียมคลอไรด์ 0-25% และเจริญได้ดีที่เกลือโซเดียมคลอไรด์เข้มข้น 5-10% มีลำดับนิวคลีโอไทด์ของ 16S rRNA gene เหมือนกับ Tetragenococus halophilus ATCC 33315 99.0% จึงระบุได้ว่าแบคทีเรียกรดแล็กติกทั้ง 2 ไอโซเลทนี้เป็นชนิด T. halophilus อย่างไรก็ตามทั้ง 2 สายพันธุ์มีความแตกต่างในการผลิตเอนไซม์ และแบบแผนการหมักน้ำตาลชนิดต่างๆ รวมถึงความสามารถในการเจริญที่อุณหภูมิ 45 องศาเซลเซียส เมื่อเลี้ยงแบคทีเรียกรดแล็กติกทั้ง 2 สายพันธุ์ (T. halophilus MS33 และ T. halophilus MCD10-5-10) ในอาหาร Fish broth ที่มีโซเดียมคลอไรด์เข้มข้น 25% และวิเคราะห์องค์ประกอบของสารระเหยด้วยวิธี Purge and trap ควบคู่กับการแยกสารและระบุชนิดของสารด้วยเทคนิค Gas chromato-graphy-Mass spectrometry (GC-MS) พบว่าสามารถสร้างสารประกอบแอลกอฮอล์ ได้แก่ Isopropyl alcohol, 1-Propanol, 1-Butanol, 1-Penten-3-ol, 3-Methyl-1-butanol, 1-Pentanol และ 1-Hexanol แบคทีเรียกรด แล็กติก T. halophilus MS33 สร้าง 2-Methylpropanal ในปริมาณที่มากกว่าตัวอย่างควบคุม (p<0.05) สายพันธุ์ MS33 นี้มีแนวโน้มสร้างสารระเหยที่ให้กลิ่นเนื้อกับผลิตภัณฑ์หมักที่ปริมาณเกลือสูง 25% และไม่สร้าง Dimethyl disulfide ซึ่งทำให้เกิดกลิ่นอุจจาระ การเร่งกระบวนการหมักน้ำปลาด้วยเอนไซม์ทางการค้าร่วมกับการใช้แบคทีเรียกรดแล็กติกคือ T. halophilus MCD 10-5-10 และ T. halophilus MS33 โดยการย่อยปลากะตักด้วยเอนไซม์อัลคาเลส (Alcalase 2.4L) ในระดับ 0.25% และเอนไซม์เฟโวไซม์ (Flavouzyme 500L) ในระดับ 0.5% น้ำปลาที่ได้มีมาตรฐานเทียบเท่ากับน้ำปลาชั้นที่ 1 หลังจากการหมักเป็นระยะเวลา 6 เดือน พบว่าจำนวนแบคทีเรียลดลงจากวันเริ่มต้น 3-4 Log CFU/ มิลลิลิตร ในเดือนที่ 2 และไม่สามารถตรวจนับแบคทีเรียได้ในเดือนที่ 3 แต่อย่างไรก็ตามปริมาณกลุ่มแอลฟาอะมิโนที่เติมกล้าเชื้อ T. halophilus มีค่าสูงกว่าตัวอย่างควบคุม โดยอยู่ในช่วง 704-727 มิลลิโมลาร์ (p<0.05) ตัวอย่างน้ำปลาที่เติมกล้าเชื้อ T. halophilus MS33 และ T. halophilus MCD10-5-10 มีปริมาณฮีสตามีนต่ำกว่าตัวอย่างควบคุม (p<0.05) ตรวจไม่พบ Dimethyl disulfide ในตัวอย่างการเติมกล้าเชื้อ T. halophilus MCD10-5-10 ในน้ำปลา จากผลการทดสอบความชอบ กลิ่น และการยอมรับรวมของน้ำปลาที่เติมกล้าเชื้อแบคทีเรียกรดแล็กติกไม่ต่างจากตัวอย่างทางการค้าที่หมักเป็นเวลา 12 เดือน (p>0.05) ทดสอบการหมักน้ำปลาด้วยการเหนี่ยวนำการย่อยสลายตัวเองของปลากะตักร่วมกับการใช้กล้าเชื้อแบคทีเรีย T. halophilus MCD10-5-10 และ T. halophilus MS33 โดยบ่มปลากะตักที่อุณหภูมิ 40 องศาเซลเซียส และเกลือโซเดียมคลอไรด์เข้มข้น 10% เป็นเวลา 4 ชั่วโมง ก่อนกระบวนการหมักน้ำปลา พบว่าการเติมกล้าเชื้อทั้ง 2 ไอโซเลทในตัวอย่างที่ผ่านการบ่มสามารถเร่งการย่อยสลายในช่วง 3 เดือนแรกของการหมัก และเมื่อหมักเป็นเวลา 7 เดือน ตัวอย่างที่ผ่านการบ่มก่อนการเติมกล้าเชื้อ T. halophilus MS33 และตัวอย่างควบคุมที่ไม่ผ่านการเหนี่ยวนำการย่อยสลายตัวเองมีปริมาณกลุ่มแอลฟาอะมิโนสูงสุด (p<0.05) ตัวอย่างที่ไม่ผ่านการเหนี่ยวนำการย่อยสลายตัวเองและเติมกล้าเชื้อ T. halophilus MCD10-5-10 มีปริมาณกลุ่มแอลฟาอะมิโนน้อยกว่าตัวอย่างควบคุม (p<0.05) หากไม่มีการใช้กล้าเชื้อการบ่มปลาก่อนการหมักไม่ได้ช่วยเร่งกระบวนการหมัก ปริมาณฮีสตามีนในทุกตัวอย่างอยู่ในช่วง 12.91-16.22 มิลลิกรัม/100 มิลลิลิตร ซึ่งเป็นค่าที่ไม่เกินค่ามาตรฐาน 20 มิลลิกรัม/100 มิลลิลิตร ดังนั้นการหมักน้ำปลาด้วยการเติมกล้าเชื้อแบคทีเรียกรดแล็กติกและเหนี่ยวนำการย่อยสลายตัวเองของปลากะตักก่อนการหมักเป็นอีกทางเลือกหนึ่งที่สามารถเร่งกระบวนการหมักน้ำปลาได้ Abstract Overall objective of this research was to develop a process that can reduce fermentation time using starter culture in conjunction with commercial enzyme or fish endogenous proteinases. Optimum autolytic activity of Indian anchovy based on the extent of proteolysis and endogenous proteolytic activity was at 50°C, 10%NaCl for 4 h. Trypsin-like proteases were mainly expressible for autolytic activity. Two isolate, MS33 and MCD10-5-10, of lactic acid bacteria isolated from fish sauce fermented for 1 and 5 months were cocci with pairs/tetrads. They grew at 0-25% NaCl with optimum NaCl concentration of 5-10 %. The result of 16S rRNA gene sequences showed homology to T. halophilus ATCC 33315 at 99.0%. Thus, these isolates were identified as T. halophilus. However, both strains showed variation in enzyme production and sugar fermentation pattern as well as growth at 45°C. Two strains of lactic acid bacteria (T. halophilus MS33 and T. halophilus MCD10-5-10) were cultured in fish broth containing 25%NaCl and volatile compounds were analyzed by purge and trap equipped with Gas chromatography-Mass spectrometry (GC-MS) for identification. These isolates produced alcohols including isopropyl alcohol, 1-propanol, 1-butanol, 1-penten-3-ol, 3-methyl-1-butanol, 1-pentanol, and 1-hexanol. Isolate MS33 produced 2-methylpropanal more than the control (p<0.05). This isolate produced volatile compounds providing “meaty” note at high salt content of 25%. Dimethyl disulfide, a compound attributing to fecal note, was not detected in the sample added T. halophilus MS33. Acceleration of fish sauce fermentation using 0.25%Alcalase 2.4L and 0.5%Flavozyme 500L in combination with a starter culture of T. halophilus MCD10-5-10 and T. halophilus MS33 was investigated. The first grade fish sauce was obtained after 6 months of fermentation. Lactic acid bacterial counts of inoculated samples decreased 3-4 Log CFU/ml at 2 months of fermentation and were not detected after 3 months. α-Amino contents of 6-mo-old fish sauce samples were 704-727 mM and higher than the control (p<0.05). Addition of T. halophilus MS33 and T. halophilus MCD10-5-10 resulted in lower histamine content than the control (p<0.05). Dimethyl disulfide was not detected in the fish sauce sample added T. halophilus MCD10-5-10. Based on the liking test, odor and overall acceptance of fish sauce fermented using lactic acid bacteria cultures were not different from those of the traditionally-fermented fish sauce (p>0.05). Fish sauce fermentation using pre-autolysis in combination with a starter culture of T. halophilus MCD10-5-10 and T. halophilus MS33 was investigated. Autolysis was induced at 40°C, 10%NaCl for 4 h before fermentation. Pretreatment of autolysis and addition both of starter cultures induced degree of hydrolysis within the first 3 months of fermentation. After 7 months, the sample pretreated with autolysis and adding T. halophilus MS33 and the control sample without pre-autolysis (no starter culture added) exhibited higher α-Amino content (p<0.05). The sample without pre-autolysis in combination with T. halophilus MCD10-5-10 showed lower α-Amino content than the control (p<0.05). Pre-autolysis without starter culture did not accelerate fermentation. Histamine content of all samples ranged 12.91-16.22 mg/100 ml, which did not exceed an allowable limit of 20 mg/100 mL. Therefore, the use of lactic acid bacterial starter culture in conjunction with pre-autolysis could be an alternative means to accelerate fish sauce fermentation.
