Abstract :
การศึกษาครั้งนี้มีวัตถุประสงค์เพื่อคัดหา Plant Growth Promoting Rhizobacteria (PGPR) ที่มีประสิทธิภาพในการส่งเสริมการเจริญเติบโตของข้าวโพดฝักอ่อนพันธุ์แปซิฟิค 283 โดยการดำเนินแยกเชื้อแบคทีเรียจากตัวอย่างดินบริเวณรากข้าวโพดในจังหวัดเชียงใหม่ ลำปาง นครสวรรค์ สระบุรี และนครราชสีมา แล้วนำมาศึกษาคุณสมบัติพื้นฐาน ในการส่งเสริมการเจริญของพืช พบว่าแบคทีเรียจำนวน 153 ไอโซเลต มีความสามารถในการสังเคราะห์ฮอร์โมนพืช (Indole-3-acetic acid: IAA) จำนวน 16 ไอโซเลต ได้คัดเชื้อ 6 ไอโซเลตที่มีประสิทธิภาพสูงสุดในการส่งเสริมการเจริญของรากข้าวโพดมา คือ ทดสอบในระดับกระถาง พบว่า ได้ไอโซเลตที่มีประสิทธิภาพในการส่งเสริมการเจริญของข้าวโพดสูงสุดคือ ไอโซเลต SUT2 และ SUT3 เมื่อนำไปทดลองในระดับแปลงที่มีการจัดกรรมวิธีใช้ร่วมกับปุ๋ยเคมี โดยลดอัตราส่วนปุ๋ยเคมีลง 25% และ 50% จากอัตราส่วนที่แนะนำ พบว่าการใส่เชื้อแบคทีเรียไอโซเลต SUT3 ร่วมกับปุ๋ยเคมีที่ลดอัตราส่วนลง มีแนวโน้มทำให้ต้นข้าวโพดมีการเจริญเติบโต และให้ผลผลิตไม่แตกต่างอย่างมีนัยสำคัญทางสถิติกับการใส่ปุ๋ยเคมีตามอัตราส่วนแนะนำเพียงอย่างเดียว การจำแนกชนิดของไอโซเลต SUT3 โดยใช้การอ่านลำดับเบสบนสายดีเอนเอของยีน 16S rDNA พบว่ามีความคล้ายกับแบคทีเรียในสกุล Bacillus subtilis คำสำคัญ: PGPR, ข้าวโพดฝักอ่อน, ปุ๋ยเคมี Abstract The objective of this study was to select highly effective Plant Growth Promoting Rhizobacteria (PGPR) for maize (Pacific 283) cultivation. The PGPR were isolated from maize rhizosphere from Chiang Mai, Lampang, Nakhon Sawan, Saraburi and Nakhon Ratchasima provinces. From 153 isolates, the isolates SUT2 and SUT3 showed the highest plant growth promotion from pot experiments. The true isolates selected for further study in the field experiment were mixed with the 25% and 50% reduction from the recommended chemical fertilizer rate. The results showed that the plant yield and biomass reduced amount of chemical fertilizer mixing with isolate SUT3 of was not significantly different from these of recommended chemical fertilizer rate. In addition, the isolate SUT3 was identified as one closely related to Bacillus subtilis sp., and there was reasonable evidence to show this isolate SUT3 could reduce the amount of chemical fertilizer used in maize field.
