Abstract :
บทคัดย่อ ซิริซินที่ได้จากการสกัดรังไหมดิบจากหนอนไหม bombyxmori ด้วยกรรมวิธีการใช้ความร้อนและความดันสูง เมื่อนำมาทดสอบประสิทธิภาพการช่วยการดูดซึมโครเมียมในลำไส้หนูทดลองและการลดระดับคลอเรสเตอรอลชนิด Low density lipoprotein (LDL)พบว่าซิริซินช่วยทำให้โครเมียมดูดซึมเข้าสู่ร่างกายอย่างรวดเร็ว โดยไม่มีผลต่อระดับการกินอาหารของหนูทดลองและอัตราการเจริญเติบโตของหนู แต่น้ำหนักเฉลี่ยของหนูกลุ่มทดลองที่ได้รับโครเมียมร่วมกับซิริซินมีแนวโน้มลดลง อาจเป็นผลจากการที่ร่างกายของหนูดูดซึมโครเมียมได้ดีมากขึ้น ซึ่งจะช่วยเร่งการเผาผลาญคาร์ไฮเดรตและไขมัน ส่งผลให้มีน้ำหนักลดลงมากกว่าในกลุ่มควบคุม สอดคล้องกับน้ำหนักไขมันบริเวณหน้าท้อง (Omentum) ที่ลดลง เมื่อนำเซลล์ไขมันบริเวณหน้าท้อง (Adipose cell) ไปตรวจเปรียบเทียบกันพบว่ามีขนาดเล็กกว่าในกลุ่มควบคุมอย่างมีนัยสำคัญทางสถิติ นอกจากนี้ปริมาณของ Cholesterol และ Triglycerides ลดลงอย่างมีนัยสำคัญทางสถิติโดยที่ระดับ HDL ของหนูที่ได้รับอาหารเสริมซิริซินโครเมียมสูงกว่าหนูที่ได้รับสารอาหารเพียงอย่างใดอย่างหนึ่ง ระดับ LDL และ Triglycerides ลดต่ำลงมากกว่า 50% เมื่อเทียบกับหนูทดลองในกลุ่มควบคุม ระดับการเสริมอาหารที่เหมาะสมที่พบคือ ซิริซินเข้มข้น 10 mg/kg ร่วมกับโครเมียม 300 µg/kg สำหรับการทดสอบความเป็นพิษต่อเซลล์เพาะเลี้ยง Caco-2 ของโครเมียมและซิริซินพบว่าไม่มีผลต่อการเจริญเติบโตของเซลล์เมื่อใช้ความเข้มข้นของโครเมียมสูงถึง 8 ppm และซิริซินที่ความเข้มข้น 100 µg/ml และที่ความเข้มข้น 1600 µg/ml จำนวนเซลล์เพาะเลี้ยงลดลง 20% จะเห็นได้ว่าซิริซินมีศักยภาพในการนำมาใช้เป็นอาหารเสริมร่วมกับโครเมียม เพื่อเพิ่มประสิทธิภาพในการดูดซึมและเสริมฤทธิ์ของโครเมียม Abstract Sericin extracted from silk cocoons of Bombyxmori under high pressure and temperature was used as a feed supplement to enhance the absorption efficiency of chromium in small intestines of rats and to lower the level of serum low density lipoprotein (LDL). Results show that sericin increased the rate of chromium adsorption without affecting the amount of feed consumption and the growth rate. However, rats fed sericin-chromium had lower body weight at the end of the study. This indicates that rats in these groups absorbed higher amount of chromium than the control group and had higher metabolism rates of carbohydrates and lipids resulting in lower body weight. The results correlate well with the weight of adipose tissue and size of adipose cells than rats fed sericin-chromium had significant lower weight omentum and smaller size of adipose cells than the rats in control group. Moreover, HDL levels of rats fed sericin-chromium were significantly higher than those of the control. Whereas, LDL and triglyceride levels were 50% lower than the control group. The appropriate ratio of sericin and chromium found in this study was 10 mg/kg and 300 µ/kg, respectively. Cell toxicity was conducted in Caco-2 cells. The concentration of chromium up to 8 ppm did not affect the viability of Caco-2 cells. Sericin at the concentration of 100 µg/ml was not toxic to Caco-2 cells. However, at the concentration of 1600 µg/ml, cell viability was reduced 20%. From this study, it suggests have potential to be use as a food supplement to enhance the adsorption efficiency of chromium.
