Abstract :
บทคัดย่อ ผู้วิจัยประสบความสำเร็จในการประยุกต์ใช้เทคโนโลยีการแสดงโปรตีนบนผิวเฟจในการคัดหา ได้เพียงเส้นเปบไทด์ที่จำเพาะต่อไคติน แต่ไม่สามารถหาเปบไทด์ที่จับกับแป้งได้ ทั้งนี้เนื่องจาก ธรรมชาติของแป้งและไคตินนั้นเป็นรูพรุนสามารถจับกับเฟจแบบไม่จำเพาะเจาะจงได้เป็นจำนวนมาก จนทำให้ไม่สามารถคัดแยกเฟจที่จับอย่างจำเพาะออกมาได้ เมื่อทำการคัดหา (biopanning) ในหลอดทดลองแบบต่างๆ อย่างไรก็ตามผู้วิจัยสามารถหาวิธีในการตรึงไคตินให้อยู่บนจาน ELISA ได้ในปริมาณเพียงพอจนสำเร็จ จึงทำให้สามารถคัดหาเฟจที่แสดงเปบไทด์ที่มีขนาดความยาว ๑๒ กรดอะมิโน ที่จับกับไคตินได้ เรียกเปบไทด์นั้นว่า ChiBP จากนั้นจึงได้ทำการทดลองเพื่อแสดงความสามารถในการใช้เปบไทด์ในการทำพันธกิจของวัสดุชีวภาพ โดยได้แก่การแสดงความสามารถในการจับกับเม็ดไคตินแล้วทำให้เม็ดเหี่ยวลง การเชื่อม ChiBP เปบไทด์กับเอนไซม์ อัลคาไล ฟอสฟาเทส แล้วนำไปจับกับเม็ดไคติน และท้ายที่สุดได้แสดงให้เห็นว่าเปบไทด์ที่คัดหามาได้ สามารถชักนำให้ ไคติน ในรูปแบบ คอลอยด์ และ ไคตินสั้นขนาดหกหน่วย รวมตัวเป็นแผ่นบางเคลือบบนผิวพลาสติกได้ เมื่อให้ความร้อนที่ ๔๒ องศาเซลเซียส โดยการสังเกตจากภาพผ่านกล้องจุลทรรศน์อิเลคตรอนแบบส่องกราด ผลจากการทดลองแสดงให้เห็นถึงศักยภาพของการประยุกต์ใช้เทคโนโลยีเฟจในการคัดหาเปบไทด์ต่อเป้าหมายหลากหลายชนิด รวมทั้งเปบไทด์ที่จำเพาะต่อไคตินที่คัดหามาได้จากการทดลองนี้ สามารถนำมาประยุกต์ใช้ในเชิง นาโนเทคโนโลยีชีวภาพด้านต่างๆ ได้อย่างกว้างขวางเพราะไคตินเป็นวัสดุชีวภาพที่มีศักยภาพทางเทคโนโลยีอย่างหลากหลาย ทั้งการแพทย์ เกษตร และสิ่งแวดล้อมความสำเร็จของการแสดงได้จากการที่ผลงานวิจัยนี้ได้รับการยอมรับให้ตีพิมพ์ในวารสารวิชาการระดับนานาชาติที่ต้องผ่านการประเมินของผู้ทรงคุณวุฒิ และได้ทำการยื่นขอสิทธิบัตรของเปบไทด์ไปแล้วด้วย ABSTRACT We were able to isolate a peptide specific to chitin, but not starch. This was because of the nature of chitin and starch that are porous and can trap large amount of non-specific binder phage, after various methods of in vitro biopanning have been done. Fortunately, we were able to develop a method for immobilizing of chitin in wells of microtiter plate, therefore a chitin-binding peptide from a 12-mer random peptide library was successfully isolated. The potential of using the selected chitin targeting peptide to functionalize biomaterial were demonstrated. The synthetic chitin binding peptide (ChiBP) could bind to chitin beads and disrupt their structure. This selected peptide was successfully used to immobilize alkaline phosphatase on chitin. In addition, the peptide could induce colloidal chitin in water to form a chitin coat on the surface of plastic tubes. Scanning electron microscopy (SEM) revealed that the peptide could induce colloidal chitin and chitohexaose to form networks when the temperature was raised to 42oC. The outcome of this research clearly demonstrates that phage display technology is one of the most powerful techniques for selecting specific peptides for a wide variety of targets. In addition this peptide can be used for various applications in nanobiotechnology because chitin is a biopolymers that have applications in agricultural, medical and environmental fields. The success of this project was evident as a publication in peer-review international journal as well as an application for a national patent.
