บทคัดย่อ งานวิจัยนี้ได้ทำการสกัดจีโนมของเชื้อแบคทีเรียในทะเล Vibrio alginolyticus สายพันธุ์ 283 และการเตรียมห้องสมุดดีเอ็นเอขนาด 3-8 กิโลเบสที่เตรียมด้วยเอ็นไซม์ตัดจำเพาะ Sau3IA การตรวจหายีนไคติเนสจากห้องสมุดดีเอ็นเอโดยวิธีทางอิมมูโนวิทยาที่ใช้ anti-V. carchariae chitinase polyclonal antibodies เป็นตัวจับแบบจำเพาะพบชิ้นดีเอ็นเอขนาด 7 กิโลเบสที่โคลนเเข้าสู่พลาสมิด pBluescript II KS(-) มีส่วนของ open reading frame ขนาด 1,740 นิวคลีโอไทด์ที่ถอดระหัสให้เอ็นไซม์ไคติเนสขนาด 63 กิโลดาลตัน การวิเคราะห์โดยวิธี immunoblotting พบว่าโปรตีนที่ถูกผลิตขึ้นสามารถจับกับไคติเนสแอนติบอดีได้อย่างดีและสามารถสลายไกลคอลไคตินได้ การเปรียบเทียบลำดับของกรดอะมิโนที่ถูกถอดระหัสมาจากยีนไคติเนสจากเชื้อ V. alginolyticus พบว่ามีความเหมือนกับไคติเนส เอ จากเชื้อ V. carchariae มากที่สุดและมีความเหมือนกับไคติเนส เอ จากเชื้อ B. circulans น้อยที่สุด การแยกโปรตีนที่จับกับไคตินจากแบคทีเรีย V. alginolyticus 283 และการวิเคราะห์มวลของเพปไทด์โดยวิธี HPLC-MS พบโปรตีน 4 ตัวที่จับแบบจำเพาะกับไคติน ผลการตรวจหาชนิดของโปรตีนโดยการเปรียบเทียบเพปไทด์กับโปรตีนในฐานข้อมูลบ่งชี้ว่าโปรตีนขนาด 90 กิโลดาลตัน 65 กิโลดาลตัน 47 กิโลดาลตัน เป็นไคติเนส ในขณะที่โปรตีนขนาด 38 กิโลดาลตัน เป็นช่องพอรินที่ถูกเหนี่ยวนำด้วยน้ำตาล การทำบริสุทธิ์โปรตีนขนาด 90 และ 65 กิโลดาลตันหรือ Chi-90 และ Chi-65 ตามลำดับ ทำได้โดยวิธีทางโครมาโตกราฟฟี Sephacryl S200HR gel filtration ผลการศึกษาทางจลนพลศาสตร์พบว่าเอ็นไซม์ Chi-65 มีอัตราการเร่งปฏิกิริยา (kcat) มากกว่าเอ็นไซม์ Chi-90 อยู่ 4.5 เท่า การศึกษาผลของ pH ต่อการเร่งปฏิกิริยาพบว่าเอ็นไซม์ Chi-90 และ Chi-65 มีค่า optimum pH เหมือนกันคือที่ pH เท่ากับ 6.5 การวิเคราะห์ผลิตผลที่เกิดจากการสลายไคตินและไคโตโอลิโกแซคคาร์ไรด์โดยวิธี TLC ให้รูปแบบคล้ายคลึงกันคือเอ็นไซม์ทั้งสองไม่สามารถสลายไคโตไบโอสได้ สลายน้ำตาล G4 ให้ผลิตผลหลักคือ G2 และสลาย G6 ให้ผลิตผลสามชนิดคือ G1 G2 และ G3 ซึ่งแสดงให้เห็นว่า Chi-90 และ Chi-65 เป็นเอนโดไคติเนส และน้ำตาลผลิตผลหลักที่ได้จากการสลายไคตินคือ G2 และ G3 Abstract This research describes the isolation of genomic DNA of a marine bacterium, Vibrio alginolyticus strain 283. A 3-8 kb DNA library was constructed from the Sau3AI partial digests and chitinase expression was detected immunologically using anti-V. carchariae chitinase polyclonal antibodies as the specific probe. A DNA fragment of approximately 7 kB, which was cloned into the pBluescript II KS(-) plasmid, was proved to contain an open reading frame of 1,740 nucleotides that encodes a 63-kDa chitinase. The expressed protein reacted strongly with anti-chitinase A antibodies and was able to hydrolyze glycol-chitin substrate. The putative amino acid sequence of V. alginolyticus chitinase displayed highest identity with V. carchariae chitinase A but lowest identity to B. circulans chitinase A1. Chitin binding proteins were further isolated from V. alginoloyticus strain 283. Tryptic peptide mass analysis by HPLC-MS identified four proteins that bound specifically to chitin. Submission of mass fingerprinting data for database search identified the 90-kDa, 65-kDa, and 47-kDa proteins as chitinases. On the other hand, the 38-kDa protein was compatible with a sugar-inducible porin. The 90-kDa and 65-kDa proteins, later designated Chi-90 and Chi-65 respectively, were further purified using Sephacryl 200HR gel filtration chromatography. Kinetic study demonstrated that Chi-65 had 4.5 folds greater catalytic rate (kcat) towards pNP-[GlcNAc]2 than Chi-90. Investigation of chitinase activity as a function of pH revealed that both enzymes worked best at pH of 6.5. Product analysis by TLC displayed similar patternes in chitooligosaccharide and chitin hydrolyses, where both enzymes did not degrade chitobiose and hydrolyze G4, yielding G2 as the major product. Three reaction products (G1, G2, and G3) were detected from G6 hydrolysis, giving an indicaition that both enzymes act as endochitinases. The final products obtained from chitin hydrolysis were G2 and G3.
