Abstract :
บทคัดย่อ โครงการการพัฒนาเทคโนโลยีการแสดงโปรตีนบนผิวฟาจเพื่อการผลิตโมโนโคลนอลแอนติบอดี (monoclonal antibody) นี้ ประสบความสำเร็จเป็นอย่างดี โดยในขั้นแรกได้ทำการพัฒนาวิธีการที่สะดวก ประหยัด รวดเร็ว และมีประสิทธิภาพดีในการคัดหาแอนติบอดีจากคลังของฟาจ รวมทั้งได้ทำการสร้างเวคเตอร์ หรือ ฟาจมิด ซึ่งสำหรับใช้ในการสร้างคลังขึ้นมาใหม่ เรียกเวคเตอร์นี้ว่า pMod1 จากนั้นจึงได้ทำการสร้างคลังของฟาจโดยใช้เม็ดเลือดขาวของมนุษย์ทั้งชายและหญิงรวม 140 คน ในจังหวัดนครราชสีมา เป็นต้นแบบในการสังเคราะห์ยีนแอนติบอดีส่วนที่มีความแปรปรวนสูง โดยการสังเคราะห์เริ่มจากการสกัด mRNA แล้วสร้างเป็น cDNA ก่อนที่จะนำมาเพิ่มจำนวนเป็น DNA ของชิ้นแอนติบดีส่วนต่างๆ ด้วยวิธีการ PCR รวมทั้งสิ้น 75 ปฏิกิริยา จากนั้นจึงนำชิ้น DNA ที่เหมาะสมมารวมกันเข้าเป็น แอนติบอดีส่วน scFv ด้วยวิธีการ PCR ก่อนที่จะนำไปโคลนเข้าเวคเตอร์ pMod1 เพื่อสร้างเป็นคลังของฟาจที่สมบูรณ์ จากการวิเคราะห์พบว่าคลังของฟาจที่แสดง monoclonal antibody ของมนุษย์ ส่วน scFv ที่ได้สร้างขึ้นนั้นมีขนาดความหลากหลายสูงถึง 1.5x108 ชนิด และมีศักยภาพสูงในการนำไปคัดหา antibody ต่อ แอนติเจน (antigen) ชนิดต่างๆ ได้อย่างจำเพาะเจาะจง ทั้งที่เป็น โปรตีนบริสุทธิ์ (BSA, GST) เป็นสารโมเลกุลเล็กประเภท hapten คือสาร อะฟลาทอกซิน และสารที่มีความสลับซับซ้อนสูง ได้แก่ พิษงูเห่า และผิวเซลล์มะเร็งท่อน้ำดี ผู้วิจัยได้ตั้งชื่อคลังนี้ว่า คลัง ย่าโม๑ ซึ่งจะสามารถนำไปใช้ประโยชน์ในการศึกษาวิจัย และการประยุกต์ใช้ในทางวิทยาศาสตร์ชีวภาพด้านต่างๆได้อย่างกว้างขวางต่อไป Abstract This research project entitled Application of phage display technology for the production of monoclonal antibody has been successfully conducted. In the frst step, an effcient method for the affnity selection of phage antibody library has been established. After that, the vector or phagemid that was used for the construction of phage display antibody library in this projected has been constructed. This vector was named pMod1. The phage-displayed antibody library was constructed by using a very large antibody variable region gene repertoires derived from 140 non-immunized donors, both male and female in Nakhon Ratchasima province. The construction of the library started from the isolation of mRNA from peripheral blood lymphocyte, cDNA synthesis, and synthesis of all possible combinations of heavy and light chains using a complex set of modifed primers by 75 independent PCR reactions, before reassembling into scFv fragments by overlapping PCR. The inserts were then cloned into pMod1 vector for the generation of phage library containing 1.5x108 individual clones. This library could be used to affnity select specifc monoclonal antibodies against a wide variety of antigens including purifed protein (BSA and GST), hapten (afatoxin), and highly complex antigens, namely crude cobra venom and cholangiocarcinoma cell surface. This high quality antibody library was designated Yamo1 and has tremendous potential for a wide area of researches and applications in the future.
