Abstract :
บทคัดย่อ ปลากะตัก (Stolephorus indicus) ซึ่งเป็นวัตถุดิบหลักของการผลิตน้ำปลามีการสะสมของฮีสตามีน พิวเทรสซีน คาดาเวอรีน และ ไทรามีน ในปริมาณสูงเมื่อเกิดการเน่าเสียที่อุณหภูมิห้อง (25ซ) เป็นเวลา 16 ชั่วโมง แต่เมื่อเก็บปลาในน้ำแข็งเป็นเวลา 13 วันจนเกิดการเน่าเสีย มีเพียงปริมาณฮีสตามีนเท่านั้นที่เพิ่มขึ้น Morganella morganii คือแบคทีเรียที่สร้างไบโอจีนิกในปริมาณสูงที่คัดแยกจากปลากะตักที่เน่าเสียที่ 25ซ ซึ่งไม่เพียงแต่ผลิตฮีสตามีน แต่ยังผลิตพิวเทรสซีนและคาดาเวอรีนได้สูง ในขณะที่ Pseudomonas fluorescens คือแบคทีเรียที่คัดแยกจากปลากะตักที่เน่าเสียในน้ำแข็งเป็นเวลา 13 วันซึ่งสามารถสร้างพิวเทรสซีนในปริมาณสูง อาหารเลี้ยงเชื้อ Plate count agar (PCA) เป็นอาหารเลี้ยงเชื้อที่มีประสิทธิภาพสูงสุดในการคัดแยกแบคทีเรียที่สร้างไบโอจีนิกเอมีนจากปลากะตัก เมื่อเปรียบเทียบกับอาหารคัดแยกเชื้อ Pseudomonas (Pseudomonad isolation, PI) และอาหาร Thiosulfate Citrate bile agar (TCBS) จากผลการวิจัยพบว่าอาหารไนเวน (Niven medium) ซึ่งใช้คัดกรองแบคทีเรียที่สร้างฮีสตามีนเป็นการเบื้องต้นนั้นไม่แสดงผลบวกที่ผิดพลาด (False-positive) จากการวิเคราะห์ปริมาณไบโอจีนิกเอมีนของผลิตภัณฑ์ปลาหมักพื้นบ้านได้แก่ ปลาร้า แหนมปลา และปลาส้ม พบปริมาณไบโอจีนิกเอมีนสูงเกินค่ามาตรฐานสากล (5-10 มก./100 ก.) ในหลายตัวอย่าง ตัวอย่างปลาร้าที่ทดสอบมีค่าคาดาเวอรีนและไทรามีนสูง ปริมาณไบโอจีนิกเอมีนมีความผันแปรสูงระหว่างตัวอย่างแหนมปลา โดยพิวเทรสซีน คาดาเวอรีน และฮีสตามีน คือไบโอจีนิกเอมีนหลักที่พบในบางตัวอย่าง คาดาเวอรีนคือไบโอจีนิกเอมีนหลักที่พบในปลาส้มโดยมีค่าสูงสุด 22.83±1.13 มก./100 ก. แบคทีเรียกรดแล็กติกที่ชอบเกลือปานกลางเป็นแบคทีเรียกลุ่มเด่นที่พบในทุกตัวอย่างปลาร้า ในขณะที่ Pseudomonas และ Enterobacteriaceae เป็นแบคทีเรียกลุ่มเด่นร่วมกับแบคทีเรียกรดแล็กติกที่พบในตัวอย่างแหนมปลาและปลาส้ม แม้ว่าแบคทีเรียกรดแล็กติกจะเป็นแบคทีเรียกลุ่มเด่น แต่กลับไม่พบการสร้างไบโอจีนิกในแบคทีเรียกลุ่มนี้ แบคทีเรียที่สร้างไบโอจีนิกเอมีนที่คัดแยกจากผลิตภัณฑ์ปลาหมักเหล่านี้คือ Enterobacter aerogenes, Providencia rettgeri, M. morganii, Klebsiella ornithinolytica, และ Staphylococcus xylosus โดย E. aerogenes ที่คัดแยกจากตัวอย่างปลาส้มแสดงความสามารถในการผลิตฮีสตามีน พิวเทรสซีน และคาดาเวอรีนสูงในปริมาณ 117.62±2.10, 204.77±1.28 และ 64.49±0.44 มก./100 มล. ตามลำดับ ในอาหารเหลวมูลเลอร์ (Moller broth) ผลิตภัณฑ์ปลาดองเค็มได้แก่ ปลาอินทรีย์เค็มและปลาทูเค็ม มีปริมาณฮีสตามีน พิวเทรสซีน และคาดาเวอรีนสูงในช่วง 27.77-46.27, 22.13-23.34 , และ 112.97-155.38 มก./100 มล. ตามลำดับ แบคทีเรียที่สร้างไบโอจีนิกเอมีนได้สูงที่คัดแยกจากตัวอย่างเหล่านี้ สามารถระบุสายพันธุ์ได้เป็น Pseudomonas aeruginosa, Photobacterium damsela, และ S. xylosus โดยไบโอจีนิกเอมีนหลักที่สร้างโดยแบคทีเรียเหล่านี้ในอาหารเหลวมูลเลอร์คือ ฮีสตามีน ซึ่งสร้างได้สูงสุด 43.85±4.14 มก./100 ก. โดย S. xylosus จากการศึกษาผลของสารเติมแต่งอาหาร ไกลซีน เอธิลีนไดเอมีนเทตราอะซิติกเอซิด (อีดีทีเอ, EDTA) เกลือโซเดียมคลอไรด์ กรดแล็กติกและซิตริก ต่อการยับยั้งการเจริญและการสร้างไบโอจีนิกเอมีนของแบคทีเรียที่สร้างไบโอจีนิกเอมีนได้สูงคือ M. morganii และ E. aerogenes ซึ่งคัดแยกจากปลากะตักที่เน่าเสียและปลาส้ม ตามลำดับ พบว่าไกลซีน (5%) EDTA (0.5%) และเกลือโซเดียมคลอไรด์ (10%) ไม่มีผลทำลายแบคทีเรียทั้ง 2 ชนิด แต่มีผลลดการสร้างไบโอจีนิกเอมีน ไกลซีนที่ระดับความเข้มข้น 5% ลดการสร้างฮีสตามีนและพิวเทรสซีนของ E. aerogenes ลง 85 และ 48% ตามลำดับ ในขณะที่ลดการสร้างฮีสตามีนของ M. morganii ลงเพียง 34% สาร EDTA ที่ระดับความเข้มข้น 0.5% มีประสิทธิภาพในการลดฮีสตามีนและพิวเทรสซีนของ M. morganii ได้ดีกว่า E. aerogenes ในขณะที่เกลือโซเดียมคลอไรด์เข้มข้น 10% สามารถยับยั้งการสร้างไบโอจีนิกเอมีนของแบคทีเรียทั้งสองได้อย่างมีประสิทธิภาพ กรดแล็กติกและซิตริกที่ความเข้มข้น 1% มีผลยับยั้งการเจริญและการสร้างไบโอจีนิกเอมีนของ M. morganii และ E. aerogenes สารเติมแต่งอาหารที่ทดสอบแสดงศักยภาพในการลดการเจริญของแบคทีเรียที่สร้างไบโอจีนิกเอมีน และ/หรือลดความสามารถในการสร้างไบโอจีนิกเอมีนของแบคทีเรียเหล่านั้น Abstract Indian anchovy (Stolephorus indicus), a major raw material of fish sauce, accumulated high levels of histamine, putrescine, cadaverine, and tyramine as it underwent spoilage at ambient temperature (25C) for 16 h. But only histamine increased when anchovies were stored in ice for 13 days. Morganella morganii was a strong biogenic amine former isolated from anchovy decomposed at 25C. It produced not only histamine but also putrescine and cadaverine in high amounts. Pseudomonas fluorescens was isolated from anchovy stored in ice for 13 days and showed high putrescine-producing ability. Plate count agar (PCA) was shown to be the most effective medium for the initial isolation of biogenic-forming bacteria from decomposed anchovies as compared to selective media, namely Pseudomonad isolation (PI) and Thiosulfate Citrate bile agar (TCBS). Based on this study, Niven medium used for initial screening of histamine formers did not show a false-positive result. When various traditionally-fermented fish products, namely Pla-ra, Nham-pla, and Pla-som, were tested for biogenic amine content, high biogenic amine contents exceeding the international maximum allowable limit (5-10 mg/100 g) were found in some samples. Most of Pla-ra samples tested contained high amounts of cadaverine and tyramine. The content of biogenic amines greatly varied among Nham-pla samples with putrescine, cadaverine, and histamine being major biogenic amines in some samples. Cadaverine appeared to be a major biogenic amine detected in Pla-som with the highest amount of 22.83 ± 1.13 mg/100g. Moderately halophilic lactic acid bacteria appeared to be prevalent in all Pla-ra samples, while Pseudomonas and Enterobacteriaceae were predominantly found along with lactic acid bacteria in Nham-pla and Pla-som samples. Despite of the prevalence of lactic acid bacteria in these fermented fish products, none of them were found to produce biogenic amines. Biogenic amine-forming bacteria isolated from these products were identified as Enterobacter aerogenes, Providencia rettgeri, M. morganii, Klebsiella ornithinolytica, and Staphylococcus xylosus. In Moller broth, E. aerogenes isolated from Pla-som showed ability to produce histamine, putrescine, and cadaverine at the high level of 117.62±2.10, 204.77±1.28, and 64.49±0.44 mg/100 ml, respectively. Salted fish products, namely salted Spanish mackerel and salted mackerel, contained high amounts of histamine, putrescine, and cadaverine in the range of 27.77-46.27, 22.13-23.34 , and 112.97-155.38 mg/100 g, respectively. Bacteria identified as biogenic amine producers isolated from these products were Pseudomonas aeruginosa, Photobacterium damsela, and S. xylosus. The major biogenic amine produced by these bacteria in Moller broth was histamine with the highest amount of 43.85±4.14 mg/100g by S. xylosus. The effect of food additives including glycine, ethylenediaminetetraacetic acid (EDTA), sodium chloride, lactic and cirtic acid on inhibition and biogenic amine production of strong biogenic amine-producing bacteria, namely M. morganii and E. aerogenes isolated from decomposed anchovy and Pla-som, was investigated. Glycine (5%), EDTA (0.5%), and sodium chloride (10%) did not show bactericidal effect on both tested bacteria, but they significantly reduced their biogenic amine-forming ability. Glycine at 5% reduced histamine and putrescine formation of E. aerogenes by 85 and 48%, respectively, while lesser effect (34%) was observed in histamine formation of M. morganii. EDTA at 0.5% appeared to be more effective in reducing histamine and putrescine formation of M. morganii than E. aerogenes, while 10% NaCl effectively inhibited formation of major biogenic amines of these bacteria. Lactic and citric acid at 1% completely inhibited growth and biogenic amine formation of M. morganii and E. aerogenes. All food additives tested showed potential to reduce biogenic amine formers and/or their biogenic amine-forming ability.
