Project Project Title : การศึกษาสภาวะที่เหมาะสมในการผลิตรีคิมบิแนนท์โปรตีนในถังหมัก (The study for optimum production conditions of recombinant proteins in bioreactor)     downloaded 59 times Researcher Name : มารินา เกตุทัต-คาร์นส์ (Mariena Ketudat-Cairns) Abstract : บทคัดย่อ รีคอมบิแนนท์โปรตีน หมายถึง โปรตีนที่สร้างขึ้นในสิ่งมีชีวิตอื่นที่ไม่ใช่ต้นกำเนิดของโปรตีนนั้นๆ ทั้งนี้ ในปัจจุบันเทคโนโลยีที่เกี่ยวข้องกับการผลิตรีคอมบิแนนท์โปรตีนเป็นอีกกลุ่มงานวิจัยที่ได้รับที่ได้รับความสนใจเป็นอย่างมาก เนื่องจากความต้องการใช้โปรตีนชนิดต่างๆ ทั้งในระดับงานวิจัยและระดับอุตสาหกรรมมีแนวโน้มสูงขึ้น โดยยีสต์ Pichia pastoris เป็นเซลล์เจ้าบ้านที่ได้รับการรายงานอย่างกว้างขวางว่ามีประสิทธิภาพในการผลิตรีคอมบิแนนท์โปรตีน ดังนั้นงานวิจัยนี้จึงมุ่งเน้นการศึกษาสภาวะที่เหมาะสมต่อการผลิตรีคอมบิแนนท์โปรตีนจาก P. pastoris ในถังหมัก โดยใช้เอนไซม์เบต้ากลูโคสิเดสเป็นโปรตีนต้นแบบ ในขั้นต้นของโครงการวิจัย ได้ทำการศึกษาพีเอชที่เหมาะสมต่อการผลิตเอนไซม์เบต้ากลูโคสิเดสด้วยกระบวนการหมักแบบ batch พบว่าพีเอชต่ำกว่า 5.0 จะทำให้ทั้งอัตราการเติบโตและการผลิตเอนไซม์เบต้ากลูโคสิเดสต่ำ โดยค่าพีเอชในช่วง 5.0-5.25 เป็นช่วงพีเอชที่เหมาะสมต่อการผลิตเอนไซม์เบต้ากลูโคสิเดสมากที่สุด และเมื่อทำการเพิ่มพีเอชจนถึง 5.5 มีผลทำให้อัตราการเติบโตสูงขึ้น ในขณะที่อัตราการผลิตเอนไซม์เบต้ากลูโคสิเดสลดลง ดังนั้น จึงเลือกใช้พีเอชในช่วง 5.0-5.25 ในการศึกษาการผลิตเอนไซม์เบต้ากลูโคสิเดสด้วยกระบวนการหมักแบบกึ่งกะอย่างง่าย พบว่าเมื่อสิ้นสุดกระบวนการหมักความเข้มข้นของเซลล์และเอนไซม์เบต้ากลูโคสิเดส สูงกว่ากระบวนการหมักแบบกะ 1.6 และ 4 เท่า ตามลำดับ สำหรับการพัฒนากระบวนการหมักสู่กระบวนการหมักแบบกึ่งกะ ที่ใช้ระบบควบคุมการเติมสารอาหารแบบย้อนกลับ เมื่อสิ้นสุดการหมักทำให้ความหนาแน่นของเซลล์ และเอนไซม์เบต้ากลูโคสิเดสมีค่าสูงกว่ากระบวนการแบบ batch ประมาณ 6-7 เท่า และ 85-95 เท่า ตามลำดับ เนื่องจากระบบที่ใช้ควบคุมการเติมสารอาหารที่ใช้ในการงานวิจัยนี้มี 2 ระบบ คือ Methanol limited fed-batch (MLFB) และ Oxygen limited fed-batch (OLFB) เมื่อพิจารณาเปรียบเทียบประสิทธิภาพของทั้งสองกระบวนการ พบว่ากระบวนการแบบ OLFB เป็นกระบวนการที่สามารถประยุกต์ใช้ในการเพิ่มอัตราการถ่ายเทออกซิเจน โดยส่งผลให้ปริมาณเมธานอลที่ใช้ไปทั้งหมด ปริมาณเซลล์และเอนไซม์เบต้ากลูโคสิเดสที่ผลิตได้ มีค่าสูงกว่ากระบวนการแบบ MLFB ทั้งนี้ไม่พบว่าสภาวะจำกัดออกซิเจนในกระบวนการแบบ OLFB จะทำให้ปริมาณเซลล์ที่มีชีวิตลดลง หรือมีการสร้าง by-product ดังนั้น กระบวนการแบบ OLFB จึงเป็นอีกทางเลือกหนึ่งสำหรับการผลิตรีคอมบิแนนท์โปรตีนโดย P. pastoris Abstract Recombinant protein is a protein which produced in other organism (host) than that of its origin. Now a day, recombinant protein technology has high impact because the increase amount and type of protein required for both research and industrial uses. Pichia pastoris has been reported as an effective host for recombinant protein production. Thus this work focused on optimizing the condition for production of recombinant protein by P. pastoris in fermenter. Thai Rosewood -glucosidase was used as the model protein. At the beginning, the optimum pH for production of -glucosidase was studied in batch fermentation technique. The optimum cultivation pH was found to be between 5.0 and 5.25. When the cultivation pH increased to 5.5, cell growth rate increased but -glucosidase production rate decreased. When the cultivation pH was lower than 5.0, both the cell growth rate and the-glucosidase production rate decreased. Therefore, the pH in the range of 5.0-5.25 was chosen for -glucosidase production in the simple fed-batch culture. The final cell density and -glucosidase concentration were higher 1.6 and 4 times than in the batch culture. The fed-batch fermentation with feedback regulation was developed. The results showed about 6-7 times higher cell density and 85-95 times higher -glucosidase concentration compared to batch culture. Two feedback regulation techniques, methanol limited fed-batch (MLFB) and oxygen limited fed-batch (OLFB), were applied for the production of -glucosidase by P. pastoris, and the efficiency of them were compared. The OLFB technique was used to increase the oxygen transfer rate. This technique gave rise to higher methanol uptake, cell density and -glucosidase concentration compared to the standard MLFB technique. Furthermore, the oxygen limitation in OLFB technique did not affect the cell viability nor generated any major by-product.
