Abstract :
บทคัดย่อ การวิจัยครั้งนี้ได้ศึกษาถึงปัจจัยที่มีผลต่อการเปลี่ยนแปลงปริมาณของ CLA ในน้ำนมโคและการเพิ่มปริมาณ CLA ในน้ำนมโคโดยการเพิ่มน้ำมันพืช และ lactic acid bacteria ในอาหารโคนม โดยแบ่งออกเป็น 1 การศึกษา และ 2 การทดลองดังนี้ การศึกษาที่ 1 การศึกษาปัจจัยที่มีผลต่อการเปลี่ยนแปลงปริมาณของ Conjugated linoleic acid (CLA) ในน้ำนม โดยการสุ่มเก็บตัวอย่างน้ำนมโคจากฟาร์มมหาวิทยาลัยทุกเดือน ๆ ละ 1ครั้ง ๆ ละ 24 ตัว ในรอบ 1 ปี เพื่อบันทึกปริมาณผลผลิตนม องค์ประกอบทางเคมีของน้ำนม และ CLA ในน้ำนม และข้อมูลจำนวนให้นม (days in milk; DIM) อุณหภูมิความชื้น ความสัมพัทธ์ และปริมาณน้ำฝน การได้รับโภชนะต่าง ๆ ในอาหาร พบว่าปริมาณ CLA ในน้ำนมจะอยู่ในช่วง 2.67 4.28 g/day ปัจจัยด้านสัตว์ทดลอง ปัจจัยด้านการให้ผลผลิต ปัจจัยด้านสิ่งแวดล้อมและปัจจัยด้านอาหารสัตว์ที่นำมาศึกษามีความสัมพันธ์ค่อนข้างต่ำต่อปริมาณ CLA ในน้ำนม ยกเว้นการได้รับ linoleic acid และ linoleic acid มีความสัมพันธ์ต่อการผลิต CLA ในน้ำนมสูง (R = 0.73, 0.71 และ R = 0.53, 0.51 ตามลำดับ) และจากการศึกษาความสัมพันธ์ของตัวแปรต่าง ๆ ต่อการผลิต CLA ในน้ำนมโดยวิธีการวิเคราะห์ความถดถอยเชิงซ้อน (Multiple regression) ซึ่งได้สมการดังนี้ Y = -3.354 + 0.002X + 0.093X + (-0.786X ) + (-0.46X ) + 0.486X + 0.039X + 0.011X + 0.088X เมื่อ Y= CLA (g/day), X = DIM (day), X = Milk yield (kg/d), X = Milk protein (%), X = Milk lactose (%), X = Total solid (%), X = Temp (C), X = LA (g/d) และ X = LNA (g/d) โดยสมการดังกล่าวมีค่า R = 0.685 การทดลองที่ 1 การศึกษาผลของการเสริมน้ำมันพืชในอาหารต่อการให้ผลผลิต และการเพิ่มปริมาณ CLA ในน้ำนมของโครีดนมลูกผสม Holstein Friesian โดยจัดกลุ่มโคนม 24 ตัวมีน้ำหนักเฉลี่ย 451 45 ก.ก. จำนวนวันของการให้นม 97 41 วัน ปริมาณน้ำนมเฉลี่ย 22.9 4.6 ก.ก. ออกเป็น 3 กลุ่ม ๆ ละ 8 ตัว ได้แก่ กลุ่มที่ 1 โคนมกลุ่มควบคุม กลุ่มที่ 2 โคนมที่ได้รับอาหารที่เสริมน้ำมันทานตะวันที่ระดับ 2% ของอาหารข้น และกลุ่มที่ 3 โคนมที่ได้รับอาหารที่เสริมน้ำนมถั่วเหลืองที่ระดับ 2% ของอาหารข้นโดยวางแผนการทดลองแบบ RCBD (Randomized Complete Block Design) ผลการทดลองพบว่า การกินได้วัตถุแห้ง การกินได้โปรตีน ปริมาณน้ำนม องค์ประกอบของน้ำนมและน้ำหนักตัวที่เปลี่ยนแปลง ไม่มีความแตกต่างอย่างมีนัยสำคัญทางสถิติ (p>0.05) ในส่วนปริมาณกรดไขมันในน้ำนมพบว่า กรดไขมันC ,C และ C มีปริมาณลดลงเมื่อเปรียบเทียบกับโคนมที่ได้รับอาหารควบคุม (p<0.05) อย่างไรก็ตามกรดไขมัน C ,C ,C ,C มีปริมาณสูงขึ้นเมื่อเปรียบเทียบกับโคนมกลุ่มควบคุม (p<0.05) และในส่วนของ CLA (cis-9, trans-11 octadecadienoic) พบว่า มีปริมาณสูงขึ้นเมื่อเปรียบเทียบกับโคนมกลุ่มควบคุม อย่างมีนัยสำคัญทางสถิติ (p<0.05) การทอลองที่ 2 การศึกษาผลของการเสริมจุลินทรีย์กลุ่ม Lactic acid bacteria ในอาหารต่อการให้ผลผลิต และปริมาณ CLA ในน้ำนมของโคนมลูกผสม Holstein Friesian โดยจัดกลุ่มโคนมลูกผสมโฮลส์ไตน์ฟรีเชี่ยน 24 ตัว มีน้ำหนักเฉลี่ย 45754 ก.ก. จำนวนวันของการให้นม 9655 วัน ปริมาณน้ำนมเฉลี่ย 22.65.7 ก.ก. ออกเป็น 3 กลุ่ม ๆ ละ 8 ตัว กลุ่มที่ 1 โคนมกลุ่มควบคุม กลุ่มที่ 2 โครีดนมที่ได้รับอาหารร่วมกับ L.plantarum ที่ระดับ 1 x 10 cfu/cow/day และกลุ่มที่ 3 โครีดนมที่ได้รับอาหารร่วมกับ L. acidophilus ที่ระดับ 1 x 10 cfu/cow/day โดยที่อาหารทั้ง3 กลุ่มเสริมน้ำนมถั่วเหลืองที่ระดับ 2% โดยวางแผนการทดลองแบบ RCBD (Randomized Complete Block Design) จากการทดลองพบว่า การกินได้วัตถุแห้ง การกินได้ของโปรตีน การกินได้พลังงานสุทธิ ปริมาณน้ำนม และองค์ประกอบของน้ำนม และปริมาณ CLA ไม่มีความแตกต่าง (p>0.05) ในส่วนระดับของ pH ภายในกระเพาะหมัก ปริมาณ VFAs พบว่าปริมาณ Acetate, butyrate และ A/P ไม่มีความแตกต่าง (p>0.05) แต่ Propionate แตกต่างจากกลุ่มที่เสริม lactic acid bacteria (P<0.05) และการเสริม lactic acid bacteria ไม่มีผลต่อจำนวนของแบคทีเรียกลุ่มต่าง ๆ ในกระเพาะหมัก (p>0.05) Abstract The objectives of this study were to determine the Studies of factors affecting change in milk CLA, Effect of supplementation of plant oil in dairy cattle diet on production and CLA in milk in dairy cattle and Effect of supplementation of Lactic acid bacteria in dairy cattle diet on production and CLA in milk in dairy cattle. The present research divided into 1 study and 2 experiments. The first study was carried out to determine the Studies of factors affecting change in milk CLA. Milk sample were collected from farm University every month and a time per month. The records of milk yield, milk composition, CLA content, day in milk, temperature, humidity, rain and feed intake during March 2004 February 2005. CLA change all year round by CLA content has between 2.67-4.28 g/day. The factors of animal, production, environment and feed intake has low correlation on milk CLA. However, linoleic acid and linolenic acid intake has high correlation on milk CLA (R = 0.73, 0.71 and R2 = 0.53, 0.51 respectively). All variables were submitted to the multiple regressions with stepwise backward elimination for a variable to remain in the predictions equation. Y = -3.354 + 0.002X1 + 0.093X2 + (-0.786X3) + (-0.46X4) + 0.486X5 + 0.036X6 + 0.011X7 + 0.088X8 When, Y = CLA (g/day), X1 = DIM (day), X2 = Milk yield (kg/d), X3 = Milk protein (%), X4 = Milk lactose (%), X5 = Total solid (%), X6 = Temp (C), X7 = LA (g/d) and X8 = LNA (g/d) (R2 = 0.685) The first experiment was carried out to determine the Effect of supplementation of plant oil in dairy cattle diet on production and CLA in milk in dairy cattle. Twenty-for Crossbred Holstein-Friesian (>87.5 Holstein-Friesian), with averaging 22.9 + 4.6 kg milk yield, 97 + 41 days in milk, 451 + 45 kg body weight, were assigned into 3 treatment groups (8 cows in each group). The first group was the unsupplemented group (control), the second group was supplemented with 2% sunflower oil in the concentrate and the third group was supplemented with 2% soybean oil in the concentrate. The experimental design was a Randomized Complete Block Design (RCBD). Dry matter intake, milk yield, milk composition and live weight change were unaffected (P>0.05) by supplementation of soybean and sunflower oils. Concentrations of C6:0, C8:0 and C16:0 in milk were significantly decreased (P<0.05) while concentrations of C18:0, C18:1n9t, C18:1n9c and C18:2n6t in milk were significantly increased (P<0.05) when compared to the control group. Supplementation of the 2 plant oils resulted in increased CLA in milk when compared to the unsupplemented control group. However, there was no significant different (P>0.05) in CLA in milk between the supplementation of the 2 oils. The second experiment was conducted to investigate the Effect of supplementation of Lactic acid bacteria in dairy cattle diet on production and CLA in milk in dairy cattle. Twenty/four Crossbred Holstein-Friesian (>87.5 Holstein-Friesian), with averaging 22.6 + 5.7 kg milk yield, 96 + 55 days in milk , 457 + 54 kg body weight, were assigned into 3 treatment groups (8 cows in each group). The first group was the unsupplemented group (control), the second group was supplemented with 1 x 109 cfu of L. plantarum and the third group was supplemented with 1 x 109 cfu of L. acidophilus. All groups supplementation of soybean oil in dairy cattle diet the experimental design was a Randomized Complete Block Design (RCBD). There were on significant differences between treatment (P>0.05) in milk production, milk composition live weight change and CLA content by supplementation of lactic acid bacteria. Rumen pH Acetate, butyrate A/P ratio and population of bacteria in rumen were no significant different (P>0.05) However, Propionate in rumen fluid were significantly increased (P<0.05) when compared to the control group.
