Abstract :
บทคัดย่อ การศึกษานี้ มีจุดมุ่งหมายในการพัฒนาเครื่องหมายชีวภาพเพื่อการตรวจสอบย้อนกลับ ปลานิลจากฟาร์ม มทส. ด้วยเทคนิค DGGE ความเข้มข้น denaturant ที่ต่างกันจะสามารถแยกความแตกต่างของสารพันธุกรรมด้วยประสิทธิภาพที่ต่างกัน สภาวะที่เหมาะสมของ DGGE จึงจำเป็นสำหรับการศึกษาประชากรจุลินทรีย์ในปลา ตัวอย่างปลานิลจำนวน 5 ตัวต่อครั้งถูกเก็บมาจากบ่อเลี้ยงที่ 5 จากฟาร์ม มทส. ในช่วงเดือนมิถุนายน 2550 ถึง เดือนเมษายน 2551 พบประชากรแบคทีเรียจากตัวอย่างปลาในฤดูฝนมีค่าแปรผันอยู่ระหว่าง 1.6 x 106 ถึง 5.1 x 107 cfu g-1 ฤดูหนาว 8.9 x 105 ถึง 1.3 x 107 cfu g-1 และฤดูร้อน 6.8 x 106 to 7.5 x 107 cfu g-1 ด้วยการเลี้ยงบนอาหารเลี้ยงเชื้อ ซึ่งกว่า 80% เป็นแบคทีเรียแกรมลบ ตัวอย่างสารพันธุกรรมจากบริเวณเหงือกและลำไส้ปลาถูกสกัดและใช้เป็นแม่แบบในการเพิ่มจำนวน 16S rDNAของจุลินทรีย์ โดยใช้ไพรเมอร์ forward ที่มี GC clamp ผลิตภัณฑ์ที่ได้ถูกนำไปทดสอบการแยกที่เปอร์เซ็นต์เจลโพลีอะคริลาไมด์ (6.5%, 8.0% และ 10.0%) ความเข้มข้น denaturant (55-45%, 30-60% และ 30-70%) ด้วยความต่างศักย์ (120 โวลต์ และ 200 โวลต์) ของการรันเจล และเวลา (5 ชั่วโมง และ12 ชั่วโมง) ที่แตกต่างกัน ผลที่ได้แสดงให้เห็นว่าที่สภาวะเจลโพลีอะคริลาไมด์ 8.0% ความเข้มข้น denaturant 30-60% ความต่างศักย์ 120 โวลต์ และเวลา 12 ชั่วโมง เป็นสภาวะที่เหมาะสมที่สุดสำหรับการศึกษาในครั้งนี้ โดยสามารถแยกความแตกต่างของชิ้นดีเอนเอที่มีสารพันธุกรรมที่ใกล้เคียงกันได้ และได้ปรับเปลี่ยนสภาวะ DGGE อีกเล็กน้อย คือ ลดความต่างศักย์ที่ใช้เป็น 100 โวลต์ และเพิ่มระยะเวลาในการทดสอบเป็น 18 ชั่วโมง ซึ่งทำให้ได้ผลการทดลองที่คมชัดขึ้น และพบว่ามีแถบดีเอนเอ 3 แถบที่พบเฉพาะในตัวอย่างปลานิลจากฟาร์ม มทส. ในทุกฤดู แต่ไม่พบจากตัวอย่างปลาจากแหล่งอื่น และคาดว่าแถบที่จำเพาะนี้น่าจะใช้เป็นตัวบ่งชี้ชีวภาพถึงแหล่งที่มาของผลิตภัณฑ์ต้นตอได้ Abstract The bacterial community of Suranaree University of Technology (SUT) tilapia was studied with the aim to develop biological markers for traceability. Different bacteria have different DNA sequences that will denature at different denaturant concentrations. The conditions of Denaturing Gradient Gel Electrophoresis (DGGE) were optimized to screen the fish bacterial community. Five fish per time were sampled from SUT farm between June 2007 to April 2008. Bacterial community from fish skin surface, gill and intestine were grown on agar plates. Total viable count (TVC) of bacteria varied between 1.6 x 106 to 5.1 x 107 colony forming units (cfu) g-1 in rainy season, 8.9 x 105 to 1.3 x 107 cfu g-1 in cool season and 6.8 x 106 to 7.5 x 107 cfu g-1 in hot season. Eighty percent of the bacteria were Gram-negative bacteria. Total DNA was extracted from the fish gills and intestines and then used as template to amplify bacterial 16S rDNA using GC clamp primers. The amplified products were tested on vary percentage of polyacrylamide gel (6.5%, 8.0% and 10.0%), denaturant concentrations (55-45%, 30-60% and 30-70%), running times (5hr and 12hr) and voltages used (120V and 200V). The results indicated that, at 8.0% polyacrylamide gel, 30-60% denaturant concentration, 12hr running time and 120V voltage were the best conditions for these screening. This condition was able to separate different sequences of bacterial DNA. The DGGE condition has been modified further by decreased the voltage to 100V and increased the runtime to 18hr to obtain better results. The results showed 3 DNA bands on DGGE gel that specific only for bacterial DNA of SUT tilapia when compared to other farms. We think that these specific bands can be used as biological marker for traceability SUT tilapia samples.
